交通任督针刺法对血管性痴呆大鼠CA1区脑源性神经营养因子及学习记忆功能的影响 被引量:1

Effects of Coordinating Rendu Acupuncture on the Brain-derived Neurotrophic Factor in CA1 Region and the Learning and Memory Function in Rats with Vascular Dementia

摘要 目的:探讨交通任督针刺疗法对血管性痴呆大鼠学习记忆功能及海马CA1区脑源性神经营养因子(BDNF)表达的影响。方法:采用不同时点永久性结扎雄性SD大鼠双侧颈总动脉(改良2-VO)法复制全脑缺血模型,随机分为空白组、假手术组、模型组、针刺组、尼莫地平组;Morris水迷宫评价大鼠造模前与治疗后空间学习记忆功能;苏木精-伊红(HE)染色观察大鼠海马CA1区组织细胞形态学变化;蛋白免疫印迹法(Western Blot)检测BDNF表达。结果:治疗后,模型组定位航行潜伏期明显较空白组及假手术组延长(P<0.05),空间搜索穿越次数较空白组及假手术对照组明显减少(P<0.05);针刺组和尼莫地平组定位航行潜伏期均明显短于模型组(P<0.05),空间搜索穿越次数均明显多于模型组(P<0.05)。海马病理组织HE染色光镜下观察,模型组大量神经元变性,体积变小,胞核与胞浆界限不清,核固缩成三角形或不规则形,核仁消失,血管、神经元及神经胶质细胞周围间隙扩大,针刺组神经元细胞均可见零星损伤细胞,海马锥体细胞层细胞形态、排列基本正常。与模型组相比,针刺组和尼莫地平组BDNF表达明显增加(P<0.05或P<0.01);与空白组及假手术组相比,针刺组与尼莫地平组BDNF表达差异无统计学意义(P>0.05);结论:交通任督针刺法对Morris水迷宫定位航行潜伏期的学习能力和空间搜索穿越次数的记忆能力有一定改善作用,可提高血管性痴呆大鼠海马CA1区BDNF表达,对缺血致大鼠海马神经元细胞损伤有一定的保护作用。 Objective:To investigate the effect of Coordinating Rendu Acupuncture on learning and memory function,and the expression of brain-derived neurotrophic factor(BDNF)in CA1 region of hippocampus in rats with vascular dementia.Methods:The model of global cerebral ischemia was established by permanent ligation of bilateral common carotid arteries(modified 2-VO)at different time points in male SD rats,the rats were randomly divided into blank group,sham operation group,model group,acupuncture group,and nimodipine group.Morris water maze was used to evaluate the spatial learning and memory function of rats before and after treatment.The histomorphological changes of hippocampal CA1 region were observed by hematoxylin-eosin staining and the expression of BDNF was detected by Western Blot.Results:The navigation latency in model group was significantly longer than that in blank group and sham operation group(P<0.05),the times of spatial search traversing were significantly less than those in blank group and sham operation group(P<0.05).The average navigation latency of acupuncture group and nimodipine group was significantly shorter than that of model group(P<0.05),and the times of space search traversing were significantly more than that of model group(P<0.05).A large number of neurons in model group were denatured and reduced in size,the boundary between the nucleus and cytoplasm was unclear,the nucleus was condensed into a triangle or irregular shape,and the nucleoli disappeared,the vessels,neurons,and neuroglia spaces were enlarged.The neurons in acupuncture group were damaged sporadically,and the morphology and arrangement of hippocampal pyramidal layer were normal(P<0.05 or P<0.01).Compared with blank group and sham operation group,there was no significant difference in BDNF expression between acupuncture group and nimodipine group(P>0.05).Conclusion:Coordinating Rendu Acupuncture can improve the learning ability of Morris water maze navigation latency and the memory ability of space searching times,and can increase the expression of BDNF in hippocampal CA1 region of vascular dementia rats,it shows some protective effect on ischemia-induced injury of hippocampal neurons in rats.
