摘要
目的 探讨生附片和炮天雄在不同时间煎煮过程中酯型生物碱的含量动态变化规律。方法 采用Agilent Eclipse XDB C_(18)(250 mm×4.6 mm, 5μm)色谱柱,流动相C为乙腈-四氢呋喃混合溶液,体积比为25∶15,流动相D为0.1 mol·L^(-1)醋酸铵溶液(每100 mL加0.05 mL冰醋酸),梯度洗脱,检测6种单、双酯型生物碱的含量;采用滴定法测定总生物碱的含量。结果 两种药材中双酯型生物碱均在2 h内分解完全,生附片与炮天雄残渣中单酯型生物碱含量呈递减趋势。生附片可检测出3种单酯型生物碱与2种双酯型生物碱,炮天雄可检测出2种单酯型生物碱与1种双酯型生物碱,生附片中双酯型生物碱显著高于炮天雄水煎液。经过24 h煎煮,生附片与炮天雄粉末总生物碱溶出率分别为44.38%和52.24%。结论 附子与炮天雄煎煮后均能降低有毒成分含量,最佳煎煮时间为2 h。
Objective To investigate the dynamic changes of ester alkaloids content in raw Fupian(Aconiti Lateralis Radix Praeparata)and processed Tianxiong(Aconitum carmichaeli Debx.).Methods An Agilent Eclipse XDB C_(18) column(250 mm×4.6 mm,5μm)was used.The mobile phase C was acetonitrile-tetrahydrofuran mixture with a volume ratio of 25:15.Mobile phase D was 0.1 mol·L^(-1) ammonium acetate solution(adding 0.05 mL glacial acetic acid per 100 mL),gradient elution was used.The contents of six ester alkaloids were detected and the content of total alkaloids were determined by titration method.Results The diester alkaloids in the two medicinal materials were decomposed completely within 2 h,and the content of single-ester alkaloids in raw Fupian(Aconiti Lateralis Radix Praeparata)and the residue of processed Tianxiong(Aconitum carmichaeli Debx.)showed a decreasing trend.Three monoester alkaloids and two diester alkaloids could be detected in raw Fupian(Aconiti Lateralis Radix Praeparata),and two monoester alkaloids and one diester alkaloid could be detected in processed Tianxiong(Aconitum carmichaeli Debx.).The diester alkaloids in raw Fupian(Aconiti Lateralis Radix Praeparata)were significantly higher than those in water decoction of processed Tianxiong(Aconitum carmichaeli Debx.).After 24 h decocting,the total alkaloid dissolution rates of raw Fupian(Aconiti Lateralis Radix Praeparata)and processed Tianxiong(Aconitum carmichaeli Debx.)were 44.38%and 52.24%,respectively.Conclusion The contents of toxic components can be reduced after decocting raw Fupian(Aconiti Lateralis Radix Praeparata)and processed Tianxiong(Aconitum carmichaeli Debx.).The best decocting time is 2 hours.
