枸石山合剂的制备工艺及薄层色谱鉴别研究

摘要 目的 优选枸石山合剂的制备工艺,进行薄层色谱定性鉴别,为合剂进一步开发提供基础性依据。方法 采用低温、恒温水浸渍,破壁打浆,纤维素酶、低温酶解,制得枸杞子原汁;采用水预煮,破壁打浆法制备石柱参原汁;采用柠檬酸食盐脱色后水预煮,破壁打汁,糊化后,α-淀粉酶酶解得到山药原汁;用L9(34)正交设计,以合剂色泽、香气、组织形态和口感优选合剂的添加剂和稳定剂。采用薄层色谱法分析合剂中的枸杞子、石柱参、山药。结果 枸杞子、石柱参、山药原汁最佳配比为10∶1∶1,混合原汁70%∶甜菊糖0.02%∶柠檬酸0.06%∶黄原胶0.04%。薄层色谱中,枸石山合剂样品与对照药材供试品、对照品,在相应的位置上呈现出相同的斑点,阴性合剂没有干扰。结论 采用水为提取溶剂,破壁打浆,低温酶解法制得原汁,具有耗时短,提取效率高,操作安全简单,环保,无需高温等优点,并且适合规模化生产。 Objective To optimize the preparation technology of Goushishan Mixture(枸石山合剂)and identify it by the thin-layer chromatography(TLC)method,for providing a basic basis for further development of the mixture.Method The raw juice of Gouqizi(Lycii Fructus)was prepared by soaking in low temperature and constant temperature water,wall breaking and pulping,cellulase and low temperature enzymatic hydrolysis.The raw juice of Shizhushen(Shizhu Ginseng)was prepared by water precooking and wall breaking beating.The raw juice of Shanyao(Chinese yam)was prepared by water precooking after decolorization by citric acid and salt,wall breaking and pulping,mashing andα-amylase hydrolysis.L_(9)(3^(4))orthogonal design was used to optimize the additives and stabilizers of the mixture according to its color,aroma,texture and taste.TLC was used to analyze Gouqizi(Lycii Fructus),Shizhushen(Shizhu Ginseng)and Shanyao(Chinese yam)in the mixture.Results The best proportion of Gouqizi(Lycii Fructus),Shizhushen(Shizhu Ginseng)and Shanyao(Chinese yam)was 10∶1∶1,mixed juice 70%∶stevia sugar 0.02%∶citric acid 0.06%∶xanthan gum 0.04%.The TLC method showed that the mixture sample had the same spot at the corresponding position with the reference drug and the reference sample,and the negative solution had no interference.Conclusion The raw juice is prepared by wall breaking pulping and low-temperature enzymatic hydrolysis using water as the extraction solvent,which has the advantages of short time consumption,high extraction efficiency,safety,convenience,environmental protection,simple operation,no high temperature,etc.,and is suitable for large-scale production.
