基于脑肠菌轴研究电针与穴位埋线对食源性肥胖大鼠肠道菌群及脑肠肽的影响 被引量:1

摘要 目的:观察电针与穴位埋线对高脂饮食诱导的食源性肥胖大鼠体质量、Lee’s指数、摄食量、肠道菌群及脑肠肽(Brain-Gut Peptide,BGP)中肽YY(Peptide YY,PYY)、胆囊收缩素(Cholecystokinin,CCK)、胰高血糖素样肽-1(Glucagon-Like Peptide-1,GLP-1)和生长激素释放肽(Growth Hormone Releasing Peptide,Ghrelin)水平的影响,探讨电针与穴位埋线治疗肥胖症的过程中,对肠道菌群及脑肠肽的影响。方法:高脂饮食成功诱导肥胖大鼠模型后,随机平均分为模型组(M组)、电针组(E组)和穴位埋线组(A组),另设普通饲料喂养空白组(B组),每组8只,雌雄各4只。电针与穴位埋线均取“天枢”穴和“丰隆”穴,电针组予频率为3 Hz-8 Hz,强度为0.5-1 mA的疏密波干预30 min,1次/d,连续6 d,休息1 d,共28 d;穴位埋线组将4-0 PDO线完全置于大鼠体内,1次/14 d,共28 d。在造模前、造模后和干预后,测量并记录大鼠体质量、Lee’s指数和摄食量。干预后,采用16S rDNA扩增子测序方法检测各组大鼠肠道菌群,使用ELISA检测各组大鼠血清中PYY、CCK、GLP-1和Ghrelin的水平。结果:与模型组比较,电针组与穴位埋线组均能使DIO大鼠体质量明显减少(P<0.05),Lee’s指数显著降低(P<0.01),摄食量显著降低(P<0.01),两者之间无明显差异(P>0.05)。肠道菌群方面,与模型组比较,电针组与穴位埋线组均能让DIO大鼠肠道菌群中的厚壁菌门相对丰度减少,拟杆菌门相对丰度增加,F/B的比值降低,两者之间无明显差异(P>0.05)。并降低DIO大鼠肠道菌群物种的总数、多样性和均匀性,并且穴位埋线的效果优于电针。模型组与空白组比较,厚壁菌门显著增加,拟杆菌门显著减少(P<0.01)。电针组和穴位埋线组与模型组比较,均表现为拟杆菌门显著增加(P<0.01),电针组厚壁菌门明显减少(P<0.05),穴位埋线组厚壁菌门显著减少(P<0.01)。与空白组比较,穴位埋线组中蓝细菌明显减少(P<0.05)。与模型组比较,电针组与穴位埋线组均使DIO大鼠血清中PYY、CCK和Ghrelin水平均显著升高(P<0.01),GLP-1水平明显升高(P<0.05)。与电针组比较,穴位埋线组血清中PYY和Ghrelin水平显著升高(P<0.01),血清中CCK和GLP-1水平无明显差异(P>0.05)。结论:电针与穴位埋线均可有效降低DIO大鼠的体质量、Lee’s指数和摄食量。可能是通过对紊乱肠道菌群的良性调节,升高血清中PYY、CCK、GLP-1和Ghrelin水平,从而减少能量的摄入和吸收,达到治疗肥胖症的目的。 Objective:To observe the effects of electroacupuncture and acupoint catgut embedding on body mass,Lee’s index,food intake,intestinal flora and serum levels of PYY,CCK,GLP-1 and Ghrelin in food-borne obese rats induced by high-fat diet,and to investigate the effects of electroacupuncture and acupoint catgut embedding on intestinal flora and brain-gut peptides during the treatment of obesity.Methods:After the obesity model was successfully induced by high-fat diet,the rats were randomly divided into model group(Group M),electroacupuncture group(Group E)and acupoint catgut embedding group(Group A),and a blank group(Group B)was fed with normal diet.The electroacupuncture and acupoint catgut embedding were taken from“Tianshu”and“Fenglong”acupoints,and the group was given sparse and dense wave interventions with frequencies of 3Hz-8Hz and intensities of 0.5-1mA for 30 min,1 time per day,6 days per duration,rest 1 day between durations,for a total of 28 days;in the acupoint catgut embedding group,4-0PDO thread was placed completely in the rats,1 time per 14 days,for a total of 28 days.Body mass,Lee’s index andfood intake were measured and recorded before,after and after the modelling intervention.After the intervention,the intestinal flora of each group of rats was detected by 16Sr DNA amplicon sequencing,and the levels of PYY,CCK,GLP-1 and Ghrelin in the serum of each group of rats were measured using ELISA.Results:Compared with the model group,both the electroacupuncture group and the acupoint catgut embedding group could significantly reduce the body mass of DIO rats(P<0.05),Lee’s index(P<0.01)and food intake(P<0.01),and there was no significant difference between them(P>0.05).In terms of intestinal flora,compared with the model group,both the electroacupuncture group and the acupoint catgut embedding group could make the relative abundance of Firmicutes phylum in the intestinal flora of DIO rats decrease,the relative abundance of Bacteroidetes phylum increase,and the ratio of F/B decrease,and there was no significant difference between them(P>0.05).And the total number,diversity and homogeneity of intestinal flora species in DIO rats were reduced,and the effect of acupoint catgut embedding was better than that of electroacupuncture.The model group showed a significant increase in Firmicutes phylum and a significant decrease in Bacteroidetes phylum compared with the blank group(P<0.01).In the electroacupuncture group and acupoint catgut embedding group,compared with the model group,both showed a significant increase in Cyanobacteria phylum(P<0.01),a significant decrease in Firmicutes phylum in the electroacupuncture group(P<0.05),and a significant decrease in Firmicutes phylum in the acupoint catgut embedding group(P<0.01).Compared with the blank group,Cyanobacteria was significantly reduced in the acupoint catgut embedding group(P<0.05).Compared with the model group,both the electroacupuncture group and the acupoint catgut embedding group caused a significant increase in the serum levels of PYY,CCK and Ghrelin in DIO rats(P<0.01),and a significant increase in the level of GLP-1(P<0.05).Compared with the electroacupuncture group,the serum levels of PYY and Ghrelin were significantly higher in the acupoint catgut embedding group(P<0.01),and the serum levels of CCK and GLP-1 were not significantly different(P>0.05).Conclusion:Both electroacupuncture and acupoint catgut embedding can effectively reduce body mass,Lee’s index and food intake in DIO rats.It is possible that the purpose of treating obesity is to elevate the serum levels of PYY,CCK,GLP-1 and Ghrelin through benign regulation of disordered intestinal flora,thus reducing energy intake and absorption.
