Study on Extraction Technology and Antioxidant Activity of Tetrastigma Hemsleyanum Polysaccharide based on Response Surface Methodology

摘要 目的:研究超声波-酶法提取三叶青多糖的工艺及其抗氧化活性。方法:以单因素实验和响应面法(RSM)优化提取条件,自变量为纤维素酶用量、提取时间、超声功率和水与材料的比例;用FRAP法、DPPH法和羟自由基清除法的总抗氧化能力测定。结果:在最佳条件下,三叶青多糖的提取率得到有效提高,最大值(4.694±0.058%)与RSM的预测值吻合良好。三叶青叶多糖(THP)显示出较强的抗氧化活性,且具有剂量依赖性。结论:三叶青多糖有可能成为一种新型抗氧化剂,并可扩大其在医药中的应用。 Objective:To study the ultrasonic enzymatic extraction technology of Tetrastigma hemsleyanum polysaccharide and antioxidant activity.Methods:the extraction conditions were optimized by single factor experiment and response surface methodology(RSM).The independent variables were the amount of cellulase,extraction time,ultrasonic power and the ratio of water to material;The total antioxidant capacity was determined by FRAP method,DPPH method and hydroxyl radical scavenging method.Results:under the optimal conditions,the extraction rate of trefoil polysaccharide was effectively improved,and the maximum value(4.694±0.058%)was in good agreement with the predicted value of RSM.THP showed strong antioxidant activity in a dose-dependent manner.Results:under the optimal conditions,the extraction rate of trefoil polysaccharide was effectively improved,and the maximum value(4.694±0.058%)was in good agreement with the predicted value of RSM.Tetrastigma hemsleyanum polysaccharid was composed of mannose,glucuronic acid,rhamnose,galacturonic acid,glucose,galactose and arabinose,and glucose was dominant(26.749±0.634%);THP showed strong antioxidant activity in a dose-dependent manner.Conclusion:THP may become a new antioxidant and expand its application in medicine.
