摘要
目的探讨CCAAT增强子结合蛋白γ(CEBPG)、DNA甲基转移酶1(DNMT1)、微小核糖核酸-214-3p(miR-214-3p)在卵巢癌组织中的表达及相关性。方法取卵巢癌患者癌组织、癌旁组织标本进行免疫组化染色,Western blot法检测CEBPG、DNMT1表达,逆转录-聚合酶链式反应(RT-PCR)检测miR-214-3p表达。比较癌旁组织、癌组织中CEBPG、DNMT1、miR-214-3p表达水平;分析癌组织中CEBPG、DNMT1、miR-214-3p表达水平与卵巢癌患者病理特征的关系;分析癌组织中CEBPG、DNMT1、miR-214-3p表达的相关性;分析CEBPG、DNMT1、miR-214-3p单一及联合检测对卵巢癌预后的预测价值。结果癌组织中CEBPG、DNMT1、miR-214-3p表达水平高于癌旁组织(P<0.05)。与无远处转移、Ⅰ+Ⅱ期、高分化、预后良好患者相比,有远期转移、Ⅲ+Ⅳ期、低中分化、预后不良患者的CEBPG、DNMT1、miR-214-3p表达水平更高(P<0.05)。Spearman相关性分析结果显示,CEBPG与DNMT1表达呈正相关(r=0.427,P=0.001);CEBPG与miR-214-3p表达呈正相关(r=0.503,P=0.001);DNMT1与miR-214-3p表达呈正相关(r=0.537,P=0.001)。三者联合对卵巢癌患者预后的预测价值最高。结论CEBPG、DNMT1、miR-214-3p在卵巢癌中表达异常与卵巢癌发生发展密切相关,三者联合对卵巢癌预后具有较高的预测价值。
Objective To explore the expression and correlation of CCAAT enhancer binding proteinγ(CEBPG),DNA methyltransferase 1(DNMT1)and micro ribonucleic acid-214-3p(miR-214-3p)in ovarian cancer tissue.Methods Immunohistochemical staining was performed on cancer tissue and adjacent tissue samples from ovarian cancer patients;Western blot was used to detect the expressions of CEBPG and DNMT1,and reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the expression of miR-214-3p.The expression levels of CEBPG,DNMT1 and miR-214-3p in adjacent tissue and cancer tissue were compared;the relationship between the expression levels of CEBPG,DNMT1 and miR-214-3p in cancer tissue and pathological features of ovarian cancer patients were analyzed;the correlation between the expressions of CEBPG,DNMT1 and miR-214-3p in cancer tissue were analyzed;the predictive value of single and combined detection of CEBPG,DNMT1 and miR-214-3p for prognosis of ovarian cancer were analyzed.Results The expression levels of CEBPG,DNMT1 and miR-214-3p in cancer tissue were higher than those in adjacent tissue(P<0.05).Compared with patients with no distant metastasis,stageⅠ+Ⅱ,high differentiation and good prognosis,patients with long-term metastasis,stageⅢ+Ⅳ,low to medium differentiation and poor prognosis have higher expression levels of CEBPG,DNMT1 and miR-214-3p(P<0.05).Spearman correlation analysis results showed that CEBPG was positively correlated with DNMT1 expression(r=0.427,P=0.001);CEBPG was positively correlated with miR-214-3p expression(r=0.503,P=0.001);DNMT1 was positively correlated with miR-214-3p expression(r=0.537,P=0.001).The combination of the three has the highest predictive value for prognosis of patients with ovarian cancer.Conclusion The abnormal expression of CEBPG,DNMT1 and miR-214-3p in ovarian cancer is closely related to the occurrence and development of ovarian cancer,and their combination has high predictive value for prognosis of ovarian cancer.
