丹参饮片、水煎液、配方颗粒的HPLC指纹图谱相关性研究 被引量:3

摘要 目的 研究丹参饮片、水煎液、配方颗粒指纹图谱的相关性。方法 采用HPLC法测定17批丹参饮片、水煎液、配方颗粒的指纹图谱,分别建立三者的对照指纹图谱,并进行相似度评价,并结合化学模式识别方法评价三者HPLC指纹图谱的相关性。结果 丹参饮片、水煎液、配方颗粒的HPLC指纹图谱均确定了8个相同的共有峰,并指认了其中7个共有峰分别为丹参素、原儿茶醛、咖啡酸、丹酚酸E、迷迭香酸、紫草酸和丹酚酸B。指纹图谱相似度评价结果显示,三者对照指纹图谱间的相似度均大于0.990;Pearson相关分析、聚类分析及主成分分析结果显示,丹参水煎液与配方颗粒质量更为接近;偏最小二乘法(PLS)分析结果表明,丹酚酸E、丹参素、原儿茶醛、咖啡酸、丹酚酸B和峰8是影响指纹图谱信息分类的标志性成分。结论 丹参饮片、水煎液及其配方颗粒的化学成分基本一致,三者质量具有良好正相关性,可为丹参配方颗粒的生产全过程质量控制提供参考。 Objective To investigate the correlation between HPLC fingerprints of Danshen Decoction Pieces,Decoction and Formula Granules.Methods The fingerprints of 17 batches of Danshen Decoction Pieces,Decoction and Formula Granules were determined by the HPLC method,and the control fingerprints of the three were established respectively.Meanwhile,the similarity was evaluated,and the correlation of the HPLC fingerprints of the three was evaluated by the chemical pattern recognition method.Results The HPLC fingerprints of Danshen Decoction Pieces,Decoction and Formula Granules all identified 8 same common peaks,and 7 of them were identified as salvianolic acid,protocatechuic aldehyde,caffeic acid,salvianolic acid E,rosmarinic acid,purple oxalic acid and salvianolic acid B.The results of fingerprint similarity evaluation showed that the similarities in the three control fingerprints were all greater than 0.990;Pearson correlation analysis,cluster analysis and principal component analysis showed that the quality of Danshen decoction was closer to that of formula granules.The results of partial least squares(PLS)analysis showed that salvianolic acid E,salvianolic acid,protocatechuic aldehyde,caffeic acid,salvianolic acid B and peak 8 were the symbolic components that impacted the classification of fingerprint information.Conclusion The chemical components of Danshen decoction pieces,decoction and its formula granules are basically consistent,and the quality of the three has a good positive correlation,which can provide reference for the quality control of Danshen formula granules in the whole production process.
