摘要
目的 探讨苦参碱对结肠癌细胞株SW620增殖、侵袭能力及对TGF-β1、Smad4蛋白表达的影响。方法 采用CCK8及平板细胞克隆形成实验来研究苦参碱对SW620细胞增殖的影响,采用Transwell小室细胞运动实验来研究苦参碱对SW620细胞侵袭转移的影响,采用细胞蜡块免疫组化定量分析苦参碱对SW620细胞TGF-β1、Smad4、Ki-67及E-cadherin蛋白表达的影响。结果 苦参碱处理后结肠癌细胞株SW620的增殖及侵袭能力明显减弱,结肠癌SW620细胞中TGF-β1及Smad4蛋白表达均缺失,苦参碱对二者蛋白表达未见明显影响。结论 实验表明苦参碱能抑制结直肠癌SW620细胞的增殖侵袭,但与TGF-β1/Smad4信号通路机制可能无关。
Objective To investigate the effect of matrine on proliferation,invasion and TGF-β1,Smad4 protein expression in colon cancer cell line SW620.Methods The effect of matrine on proliferative activity in SW620 cells were evaluated by CCK8 assay and colony formation assay.The effect of matrine on migration and invasion ability in SW620 cells were evaluated by Transwell assay.Quantitative analysis the effect of matrine on Ki-67,E-cadherin,TGFβ1 and Smad4 proteins expression in SW620 cell blocks by Immunohistochemistry.Results The proliferation and invasion of SW620 cells were significantly weakened after matrine treatment.TGF-β1 and Smad4 proteins expression in SW620 cells were absent,matrine had no significant effect on the proteins expression of TGF-β1 and Smad4.Conclusion Matrine can inhibit the proliferation and metastasis of SW620 cells,but the TGF-β1/Smad4 signaling pathway mechanism may be independent.
