摘要
机械力敏感型离子通道是一类能够感知和响应机械力刺激的膜蛋白,其广泛存在于各种生物体中,包括细菌、植物和动物等。为了更好地了解植物机械力敏感型离子通道,本研究通过体外表达纯化以及去垢剂筛选等手段,获得了性质较好的拟南芥(Arabidopsis thaliana,At)机械力敏感型离子通道AtMSL10的蛋白样品,并利用冷冻电镜单颗粒重构技术解析了其高分辨率三维结构,整体分辨率为0.27 nm。结构显示AtMSL10蛋白以同源七聚体的形式存在,并处于开放状态的构象。AtMSL10蛋白结构的解析为后续对于植物机械力传导机制的探索提供了结构基础。
Mechanosensitive(MS)ion channels are one kind of membrane proteins that can sense and respond to the mechanical stimuli.They are highly conserved in all kingdom of life,including bacteria,plants and animals.To understand the mechanism of the MS ion channels in plants,we expressed the full⁃length AtMSL10 in insect cells,and obtained well⁃behaved proteins suitable for Cryo⁃EM analysis after screening multiple detergents.Finally,we determined the Cryo⁃EM structure of AtMSL10 at an overall resolution of 0.27 nm.The structure shows that AtMSL10 forms a homo⁃heptamer,and it exists in the open⁃state conformation.Our study provides a framework for further investigation of the plant mechanical force transduction mechanism.
