摘要
优化北虫草菌糠中多糖的提取工艺,测定北虫草菌糠中多糖的含量,期望为全面开发利用北虫草菌糠提供一定的理论依据。以北虫草菌糠为原料,利用水提醇法提取菌糠中的多糖,通过响应面法研究了提取时间、温度、料液比对北虫草菌糠中多糖的提取率及提取量的影响,并用1-苯基-3-甲基-5-吡唑啉酮衍生法研究了菌糠中多糖的组成及比例。北虫草菌糠中多糖的最佳提取条件是温度为55℃,料液比为1∶30,提取时间为1 h,得到多糖的提取率为13.95%,多糖含量为279 mg/kg,并提取得到12种单糖。此提取工艺可以用于后续的免疫活性物质的提取研究。
By optimizing the extraction process of polysaccharides from Cordyceps sinensis fungal bran,and determination the polysaccharides in Cordyceps sinensis mushroom bran,the research aims to provide some theoretical basis for the comprehensive development and utilization of Cordyceps sinensis cultivated chaff.Taking the spent mushroom substrate of Cordyceps sinensis as materials,polysaccharides were prepared from water extraction and alcohol precipitation method.The effects of extraction time,temperature and solid-liquid ratio on the extraction rate and amount of Cordyceps sinensis bran were studied by responsesurface method,and the composition and proportion of polysaccharides were measured by PMP derivations.The results showed that the optimum extraction conditions were 55 ℃,1∶30 liquid-solid ratio and 1 h extraction time.Under these conditions,the extraction rate of polysaccharide was 13.95%,and the concentrationof polysaccharide was 279 mg/kg.Twelve polysaccharides were obtained.The optimized extraction method can be used for subsequent studies on immunologically active substances.
