摘要
目的探讨细胞分裂周期相关基因8(CDCA8)在多种人恶性肿瘤中的表达及其与患者预后、肿瘤浸润免疫细胞的相关性。方法从癌症基因组图谱(TCGA)和基因型-组织表达(GTEx)、基因表达谱交互分析(GEPIA)、TIMER 2.0等数据库中获取数据,分析各种恶性肿瘤中CDCA8基因的表达水平,探讨CDCA8表达水平用于鉴别癌组织和正常组织的诊断价值,明确CDCA8表达水平与患者预后生存的关系,分析CDCA8表达与肿瘤浸润免疫细胞的相关性。结果与正常组织相比,在大多数人恶性肿瘤组织中CDCA8的表达显著上调(P<0.05)。综合受试者工作特征(SROC)曲线分析显示,CDCA8表达水平可用于癌组织和正常组织的鉴别。生存分析结果显示,高CDCA8组的恶性肿瘤患者具有更差的总生存期和无病生存期(Log-rank P<0.05)。基因本体论(GO)和京都基因与基因组百科全书(KEGG)分析显示,CDCA8可能与细胞周期和有丝分裂有关(P<0.05)。在TCGA数据库恶性肿瘤中,CDCA8表达水平与CD4+Th2细胞浸润明显相关(P<0.05)。结论CDCA8有望成为人恶性肿瘤诊断和预后评估的生物标志物,为临床上恶性肿瘤的诊治提供新的理论依据。
Objective To investigate the expression of cell division cycle associated 8(CDCA8)in various human malignant tumors and its correlation with prognosis and tumor infiltrating immune cells.Methods Based on the data from the Cancer Genome Atlas(TCGA),the Genotype-Tissue Expression(GTEx),Gene Expression Profiling Interactive Analysis(GEPIA),TIMER 2.0 and other databases,we analyzed the expression level of CDCA8 in various malignant tumors,the diagnostic value of CDCA8 expression level in distinguishing cancer tissues from normal tissues,and the relationship between the expression level of CDCA8 and the survival of patients.Meanwhile,the correlation between CDCA8 expression level and tumor infiltrating immune cells was determined.Results Compared with normal tissues,CDCA8 significantly upregulated in most human malignant tissues(P<0.05).Comprehensive receiver operating characteristic(SROC)curve analysis showed that CDCA8 expression levels cluld be used to distinguish malignant tumors from normal tissues.Survival analysis showed that patients with malignant tumors in the high CDCA8 group had worse overall survival and disease-free survival(Log-rank P<0.05).Gene Ontology(GO)and the Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis revealed that CDCA8 might be involved in cell cycle and mitosis(P<0.05).In TCGA database malignancies,CDCA8 expression level was significantly associated with CD4+Th2 cell infiltration(P<0.05).Conclusion CDCA8 is expected to become a biomarker of diagnosis and prognosis,which might provide a new theoretical basis for the diagnosis and treatment of malignant tumors.
