摘要 为提高T-2毒素降解酶产生菌Bacillus sp.AFJ-3的产酶量,同时降低发酵成本,试验以T-2毒素降解酶酶活力为评价指标,通过单因素试验、Plackett-Burman试验和Box-Behnken试验对其发酵条件和培养基进行优化。结果表明,最佳产酶发酵工艺为酵母提取物5 g/L,MgSO_(4)·7H_(2)O 16 g/L,黄豆饼粉5 g/L,培养基初始pH 5.8,温度36.5℃,接种量0.5%,转速200 r/min。在此优化条件下,T-2毒素降解酶活力达到1674.68 U/mL,较优化前提高了47.39%。研究结果可为T-2毒素降解酶的分离纯化与酶制剂的开发应用提供参考依据。 The purpose of this study was to increase the enzyme production of T-2 toxin-degrading enzyme produced by bacteria Bacillus sp.AFJ-3,and to reduce the fermentation costs.Using T-2 toxindegrading enzyme activity as an evaluation indicator,the fermentation conditions and media were optimized by single-factor,Plackett-Burman and Box-Behnken test.As a result,the optimal enzymeproducing fermentation process was yeast extract 5 g/L,MgSO_(4)·7H_(2)O 16 g/L,soybean powder 5 g/L,media initial pH 5.8,temperature 36.5℃,inoculation 0.5%,and rotation speed 200 r/min.Under this optimized condition,the enzymatic activity of T-2 toxin-degrading enzyme reached 1674.68 U/mL,which was 47.39%higher than that before optimization.The results provide a reference for the isolation and purification of T-2 toxin-degrading enzymes and the development and application of enzyme preparations.
