摘要
目的制备鞣花酸纳米微囊并观察其对乳腺癌MCF-7细胞株行为学特征的影响, 以分析其抗肿瘤效果有无改进。方法采用层层自组装法制备鞣花酸壳聚糖纳米微囊, 观察其体外抗氧化活性与游离鞣花酸的差异性。同时体外培养MCF-7细胞, 设立MCF-7组、游离鞣花酸组和纳米微囊组(均n=3), MCF-7组正常培养, 游离鞣花酸组和纳米微囊组分别加入游离鞣花酸和鞣花酸纳米微囊共培养48 h, 采用细胞计数试剂盒(CCK-8)实验与流式细胞实验检测细胞增殖与凋亡活力。计量资料先行正态分布和方差齐性检验, 计数资料以率和构成比表示, 两组间比较采用卡方检验。结果抗氧化测试结果显示, 鞣花酸纳米微囊的吸光度(A734)值为0.652±0.134, 明显低于游离鞣花酸的0.855±0.121(P<0.05)。体外细胞学实验结果显示, MCF-7组、游离鞣花酸组、纳米微囊组的A450值依次为0.854±0.102、0.595±0.077、0.452±0.046, 组间差异有统计学意义(F=20.265, P<0.01), 且纳米微囊组高于游离鞣花酸组(F=12.258, P<0.05), 游离鞣花酸组高于MCF-7组(F=31.024, P<0.05);MCF-7组、游离鞣花酸组、纳米微囊组的细胞凋亡率依次为(6.25±0.87)%、(15.63±2.22)%、(21.52±2.54)%, 组间差异有统计学意义(F=43.985, P<0.01), 纳米微囊组低于游离鞣花酸组(F=26.071, P<0.05), 且游离鞣花酸组低于MCF-7组(F=14.356, P<0.01)。结论成功制备了鞣花酸纳米微囊, 并证实其体外抗氧化活性和抗乳腺癌活性优于游离的鞣花酸。
Objective To prepare an ellagic acid nano supplementation and observe its effect on the behavioral characteristics of MCF-7 cell lines,so as to analyze whether its anti-tumor effect has been improved.Methods Chitosan nano microcapsules were prepared by layered self-assembly method,and the difference between antioxidant activity and free ellagic acid in vitro was observed.At the same time,MCF-7 cells were cultured in vitro,including MCF-7 group,free ellagic acid group and nano microcapsule group(n=3).MCF-7 group was cultured normally,free ellagic acid group and nano capsule group were co-cultured with free ellagic acid and ellagic acid nano microcapsule for 48 h,respectively.Cell counting kit-8(CCK-8)and flow cytometry experiments were used to detect cell appreciation and apoptosis viability.The counting data were expressed by rate and constituent ratio.The comparison between the two groups was performed by Chi-squared test.Results The antioxidant test results showed that the absorbance(A)734 of ellagic acid nano supplementation was 0.652±0.134,which was significantly lower than that of 0.855±0.121 of free ellagic acid(P<0.05).The results of in vitro cytology experiments showed that the A450 values of MCF-7 group,free ellagic acid group and nano supplementation group were 0.854±0.102,0.595±0.077 and 0.452±0.046 respectively,with the differences between the groups being significant(F=20.265,P<0.01),and those in the nano supplementation group were higher than in the free ellagic acid group(F=12.258,P<0.05),and those in the free ellagic acid group was higher than those in the MCF-7 group(F=31.024,P<0.05).The apoptosis rates in MCF-7 group,free ellagic acid group and nano supplementation group were(6.25±0.87)%,(15.63±2.22)%,and(21.52±2.54%)%respectively,with the differences between the groups being significant(F=43.985,P<0.01),those in the nano supplementation group was lower than in the free ellagic acid group(F=26.071,P<0.05),and those in the free ellagic acid group was lower than in the MCF-7 group(F=14.356,P<0.01).Conclusion The ellagic acid nano microcapsule was successfully prepared,and it was confirmed that its antioxidant activity and breast cancer activity in vitro were better than those of free ellagic acid.
