BvHIPP24介导甜菜应答镉胁迫的作用分析

摘要 为探究甜菜重金属相关异戊二烯化植物蛋白24(BvHIPP24)在重金属镉胁迫下的作用机制,本研究利用生信方法预测了该基因上游的顺式作用元件、转录因子和下游的互作蛋白,并分析了它们在镉胁迫下的转录表达特性。结果表明,BvHIPP24含有CGTCA-motif、生长素类、光响应、MYB结合、厌氧诱导和水杨酸应答相关的作用元件;转录因子BVRB_2g046790、BVRB_1g021800和BVRB_6g128880可能参与BvHIPP24的转录调控;BvHIPP24的互作蛋白主要包括SOD、NaKR1-like、COX1、MT2、NaKR3、REP1、RCA和LHCP等,它们可能通过与重金属离子结合参与离子运输和维持细胞内环境稳态。转录组测序结果表明BvHIPP24及互作蛋白的转录表达均受到镉胁迫的调控。综上推测,BvHIPP24主要通过上游转录因子的调控表达,直接或间接的与互作蛋白共同介导镉离子螯合及解毒。本研究为进一步揭示甜菜对镉胁迫的响应机制提供了参考。 The aim was to explore the mechanism of heavy metal-related isoprenoid plant protein 24(BvHIPP24)in sugar beet under cadmium stress.This study used bioinformatics methods to predict the cisacting elements,transcription factors,and interacting proteins upstream of the gene,and analyzed their transcriptional expression characteristics under cadmium stress.The results showed that BvHIPP24 contained cis-acting elements related to CGTCA-motif,auxin response,light response,MYB binding,anaerobic induction,and salicylic acid response.Transcription factors such as BVRB_2g046790,BVRB_1g021800 and BVRB_6g128880 might be involved in the transcriptional regulation of BvHIPP24.The interaction proteins of BvHIPP24 mainly included SOD,NaKR1-like,COX1,MT2,NaKR3,REP1,RCA and LHCP,and etc.,which might participate in ion transport and intracellular homeostasis by binding with heavy metal ions.Transcriptome sequencing results indicated that the transcriptional expression of BvHIPP24 and its interacting proteins were regulated by cadmium stress.Based on the above findings,BvHIPP24 was mainly expressed through the regulation of upstream transcription factors,and directly or indirectly mediated cadmium ion chelation and detoxification together with interacting proteins.This study provided a reference to further uncover the response mechanisms of sugar beets to cadmium stress.
