冻—放射复合伤中雄性小鼠生殖系统损伤情况及其机制研究

Study on reproductive system injury and its mechanism in male mice induced by cold-radiation

摘要 目的探讨冻—放射复合伤中小鼠生殖系统损伤情况及其可能的损伤机制。方法选取SPF级6周龄雄性BALB/c小鼠32只,体质量(20.19±1.42)g,体质量范围为18~22 g。将32只雄性小鼠随机分成对照组、寒冷组、放射组和冻—放射组,每组8只。所有小鼠适应性饲养7 d,其中寒冷组和冻—放射组小鼠每日置于人工气候箱内8 h,箱内温度设置为-10℃,8 h后返回恒温动物房,共持续7 d。第8天放射组和冻—放射组小鼠接受X射线。观察所有小鼠的一般情况,计算睾丸脏器指数,检测小鼠的精子质量和睾酮、卵泡刺激素(FSH)和黄体生成素(LH)的水平。结果冻—放射组小鼠睾丸脏器指数与对照组比较,差异无统计学意义(P>0.05)。冻—放射组精子密度[(27.01±6.15)106/ml]与精子活力[(40.22±5.71)%]较对照组显著降低[(35.80±3.41)106/ml、(62.28±2.10)%],差异有统计学意义(P<0.05)。冻—放射组精子畸形率[(20.56±4.55)%]较对照组显著升高[(10.43±2.05)%],差异有统计学意义(P<0.05)。冻—放射组睾酮水平[(28.09±5.58)nmol/L]较对照组[(39.25±1.50)nmol/L]显著降低,差异有统计学意义(P<0.05),冻—放射组FSH水平[(80.24±6.50)IU/L]较对照组[(43.11±2.86)IU/L]显著升高,差异有统计学意义(P<0.05),冻—放射组LH水平[(68.52±7.62)μg/L]较对照组[(150.58±12.30)μg/L]显著降低,差异有统计学意义(P<0.05)。结论冻—放射复合伤可导致小鼠精液参数下降、生育力受损伤,寒冷与辐射可影响小鼠血清性激素水平。 Objective To investigate the injury of reproductive system in mice with cold-radiation combined injury and its possible mechanism.Methods A total of 32 SPF male BALB/c mice,weighing(20.19±1.42)g,ranged from 18 to 22 g,were randomly divided into control group,cold group,radiation group and cold-radiation group,with 8 mice in each group.All the mice were adaptively fed for 7 d.The mice in the cold group and the cold-radiation group were placed in an artificial climate chamber for 8 hours a day,and the temperature was set to-10℃.On day 8,the mice in the radiation and cold-radiation groups received X-rays.The general condition of all mice was observed,the testis organ index was calculated,the sperm quality and the levels of testosterone,follicle stimulating hormone(FSH)and luteinizing hormone(LH)were detected.Results There was no significant difference in the testis organ index between the cold-radiation group and the control group(P>0.05).The sperm density[(27.01±6.15)106/ml]and sperm motility[(40.22±5.71)%]in the cold-radiation group were significantly lower than those in the control group[(35.80±3.41)106/ml,(62.28±2.10)%],the differences were statistically significant(P<0.05).The rate of sperm abnormality in the cold-radiation group[(20.56±4.55)%]was significantly higher than that in the control group[(10.43±2.05)%],the difference was statistically significant(P<0.05).The level of testosterone in the cold-radiation group[(28.09±5.58)nmol/L]was significantly lower than that in the control group[(39.25±1.50)nmol/L],the difference was statistically significant(P<0.05).The level of FSH in the cold-radiation group[(80.24±6.50)IU/L]was significantly higher than that in the control group[(43.11±2.86)IU/L],and the difference was statistically significant(P<0.05).The level of LH in the cold-radiation group[(68.52±7.62)μg/L]was significantly lower than that in the control group[(150.58±12.30)μg/L],the difference was statistically significant(P<0.05).Conclusions Cold and radiation combined injury can lead to decreased semen parameters and impaired fertility in mice,cold and radiation can affect serum sex hormone levels in mice.
