摘要
【目的】探究油榄仁组培快繁各阶段的最佳培养基配方以及硅酸钾对苗高整齐度的影响,建立油榄仁高质量组培快繁体系,为油榄仁良种繁育和定向培育提供理论和技术支持。【方法】以油榄仁优良家系种子为试验材料,开展消毒方法以及增殖和生根阶段培养基配方筛选试验,探讨硅酸钾对增殖苗整齐度的调控方法,分析不同处理间种子消毒效果、增殖质量以及生根情况的差异,确定适宜的消毒方法以及增殖和生根培养基。【结果】合适的种子消毒方法为:油榄仁种子经浓硫酸浸泡7 h后,用0.1%升汞溶液消毒种胚8 min,无菌胚萌芽率超过80%。增殖培养中,6-BA与IBA质量浓度比值为10,且KH_(2)PO_(4)和MgSO_(4)·7H_(2)O的质量浓度分别为MS培养基的2倍和1.5倍时,油榄仁的增殖效果最好,表现为增殖系数高且生长状态好,接种培养30 d其增殖系数、增殖苗高度和叶绿素含量分别可达5.6、3.75 cm和2.51 mg·g-1。油榄仁的最佳增殖培养基为改良MS(KH_(2)PO_(4)和MgSO_(4)·7H_(2)O的质量浓度分别为255 mg·L^(-1)和740 mg·L^(-1))+1.0 mg·L^(-1)6-BA+0.1 mg·L^(-1)IBA+30 g·L^(-1)蔗糖。硅酸钾可以提高增殖苗的整齐度,其最佳浓度为1.5 mmol·L^(-1),整齐度比对照高2.63倍。通过生根培养筛选出最佳生根配方为WPM+0.2 mg·L^(-1)NAA+20 g·L^(-1)蔗糖,生根率达94%以上。【结论】构建了油榄仁的组培快繁体系,尤其是通过培养基中添加适宜浓度的硅酸钾可改善油榄仁组培苗的整齐度。该体系不仅可获得优质组培苗,还能提高组培苗的整齐度,降低育苗成本,为油榄仁优良种质组培苗产业化生产提供技术支撑。
【Objective】The purpose of this study was to explore the optimal medium components at each stage of tissue culture and the effect of potassium silicate on the height uniformity of plantlets,and a high-quality tissue culture system of Terminalia bellirica was established,so as to provide theoretical and technical support for the breeding and targeted cultivation of T.bellirica.【Method】Using seeds of the elite T.bellirica family as the material,the optimal disinfection method and suitable medium components for proliferation and rooting were screened,and the regulation mechanism of potassium silicate on plantlet uniformity was explored.The suitable seed disinfection methods and proliferation and rooting media components were determined by analyzing differences in seed disinfection effect,proliferation quality and rooting situation among different treatments.【Result】The proper seed disinfection method was soaking the seeds with concentrated sulfuric acid for 7 h and sterilizing the seed embryos with 0.1%mercury solutions for 8 min.In this way,the germination rate of aseptic embryos could exceed 80%.In the proliferation culture,when the ratio of 6-BA to IBA was 10,and the concentrations of KH_(2)PO_(4)and MgSPO_(4)·7H_(2)O were 2 times and 1.5 times that of MS medium,respectively,the proliferation effect of T.bellirica was the best,with a high proliferation coefficient and good growth state.The proliferation coefficient,plantlet height and chlorophyll content could reach up to 5.6,3.75 cm and 2.51mg·g-1FW,respectively,after 30 days of inoculation.The optimal proliferation medium for T.bellirica was selected as a modified MS medium containing 1.0 mg·L^(-1)6-BA,0.1 mg·L^(-1)IBA,30 g·L^(-1)sucrose,255 mg·L^(-1)KH_(2)PO_(4)and 740 mg·L^(-1)MgSPO_(4)·7H_(2)O.In addition,we found that potassium silicate could improve the uniformity of the proliferated plantlets,and its optimal concentration was 1.5 mmol·L^(-1).The uniformity was 2.63 times higher than that of the control.The best medium for rooting was the Wood Plant Medium supplemented with 0.2 mg·L^(-1)NAA and 20 g·L^(-1)sucrose,and the rooting rate could be above 94%.【Conclusion】The tissue culture system of T.bellirica was completely established in the present study.In particular,it is the first report on improving the uniformity of T.bellirica plantlets through adding suitable concentrations of potassium silicate into the proliferation medium.The system can not only obtain high-quality plantlets with high height,chlorophyll content and proliferation coefficients,but also improve the uniformity of proliferated plantlets,and reduce the cost for plantlet production,which provides technical support for tissue culture and micro-propagation of the elite T.bellirica germplasm.
