摘要
目的:利用基因芯片数据库对腰椎间盘突出症(LDH)基因探针芯片数据进行富集分析和免疫浸润分析,为深入了解LDH的免疫相关机制提供理论依据。方法:运用R语言及相关安装包程序对LDH差异基因进行KEGG富集分析;通过CIBERSORT反卷法对22类免疫细胞在LDH的含量及比例进行分析;利用COREMINE数据库对显著富集的免疫相关生物学过程及核心靶基因进行中药预测。结果:LDH相关的蛋白网络互作(PPI)核心靶基因涉及PLP1、CD86、NRCAM、FCGR1A、MPO、KIF1A等15个基因。KEGG富集分析显示差异基因与Th17细胞分化、T细胞受体信号通路及炎症相关途径密切相关。LDH免疫细胞的浸润矩阵分析结果表明,巨噬细胞、肥大细胞、调节性T细胞等免疫细胞与正常组存在差异。免疫细胞间相关性分析显示,γδT细胞与滤泡辅助性T细胞呈正相关关系,中性粒细胞与单核细胞呈负相关关系。COREMINE预测显示,三七、人参、生地黄、枸杞与LDH相关免疫途径及核心靶基因的关系最为密切。结论:巨噬细胞、调节性T细胞与LDH的免疫浸润关系密切,LDH相关的差异基因涉及多种免疫相关生物学过程及通路,筛选出的补气养血药、益肝肾药、补骨髓药及活血化瘀药等中药可能作为其潜在的分子药物来源。
Objective:With the Gene Expression Omnibus(GEO)database,enrichment analysis and immune infiltration analysis were performed on lumbar disc herniation(LDH)gene probe chip data to provide a theoretical basis for understanding the immune-related mechanisms of LDH.Methods:R language and related package programs were used to perform KEGG pathway enrichment analysis on LDH differentially expressed genes.CIBERSORT was used to analyze the content and proportion of 22 types of immune cells in LDH.The COREMINE database was used to predict traditional Chinese medicines related to significantly enriched immune-related biological processes and core target genes.Results:The LDH-related protein-protein interaction(PPI)core target genes involved 15 genes,including PLP1,CD86,NRCAM,FCGR1A,MPO,KIF1A,etc.KEGG pathway enrichment analysis showed that LDH was closely related to Th17 cell differentiation,T cell receptor signaling pathway,and inflammation-related pathways.Analysis of the immune cell infiltration matrix of LDH showed differences in immune cells such as macrophages,mast cells and regulatory T cells(Tregs),compared to the control group.In the correlation analysis of immune cells,γδT cells of LDH showed strong positive correlation with follicular helper T cells,while neutrophils showed strong negative correlation with monocytes.Through COREMINE prediction,Sanqi(Panax notoginseng),Renshen(Ginseng),Shengdihuang(Rehmannia glutinosa)and Gouqi(Lycium barbarum)were found to have the closest relationship with LDH-related immune pathways and core target genes.Conclusion:The immune infiltration of macrophages and regulatory T cells is closely related to LDH.LDH-related differentially expressed genes are involved in various immune-related biological processes and pathways.Prediction Chinese medicines,including qi tonifying and blood nourishing medicine,liver and kidney tonifying medicine,nourishing bone marrow medicine,and promoting blood circulation to remove blood stasis,etc,may serve as potential molecular drug sources for LDH.
