摘要
目的 研究紫白膏对大鼠肛周创面愈合及转化生长因子β1(TGF-β1)、Smad3表达的影响。方法将40只SD大鼠随机分为空白组、模型组、对照组和紫白膏组,每组10只。除空白组外,其余3组行大鼠肛周创面造模。模型组不予特殊治疗,紫白膏组、对照组分别予紫白膏和凡士林外敷干预,均连续干预14 d。观察4组大鼠治疗14 d后的创面愈合率;采用Western blot和qPCR检测大鼠创面组织中TGF-β1、Smad3蛋白及mRNA表达情况。结果 与空白组比较,模型组大鼠肛周创面愈合率明显降低(P<0.05),创面组织TGF-β1、Smad3蛋白表达量和mRNA相对表达水平均明显提高(P均<0.05);与模型组比较,紫白膏组和对照组大鼠肛周创面愈合率均明显提高(P均<0.05),创面组织TGF-β1、Smad3蛋白表达量和mRNA相对表达水平均明显降低(P均<0.05);与对照组比较,紫白膏组大鼠肛周创面愈合率均明显提高(P<0.05),创面组织TGF-β1、Smad3蛋白表达量和mRNA相对表达水平均明显降低(P均<0.05)。结论 紫白膏可能通过调控大鼠创面组织TGF-β1、Smad3的表达来促进肛周创面愈合。
Objective:To study the effect of Zibai ointment on perianal wound healing and the expression of transforming growth factor-β1(TGF-β1)and Smad3 in rats.Methods:A total of 40 SD rats were randomly divided into blank group,model group,control group and Zibai ointment group,with 10 rats in each group.Except for the blank group,the remaining three groups were subjected to rat perianal wound modeling.Rats in the blank and model groups were given physiological saline gauze for wet application;the control group was given vaseline for external application;the Zibai ointment group was given Zibai ointment for external application.All groups were continuously intervened for 14 days.The wound healing rate was calculated after 14 days of treatment.Western blot and qPCR were used to detect the protein and mRNA expression of TGF-β1 and Smad3 in rat wound tissue.Results:Compared with the blank group,the wound healing rate in the model group significantly decreased(P<0.05),and the protein and mRNA expression of TGF-β1 and Smad3 in rat wound tissue significantly increased(P<0.05).Compared with the model group,the wound healing rate in the Zibai ointment and control groups significantly increased(P<0.05),and the protein and mRNA expression of TGF-β1 and Smad3 in rat wound tissue significantly decreased(P<0.05).Compared with the control group,the wound healing rate in the Zibai ointment group significantly increased(P<0.05),and the protein and mRNA expression of TGF-β1 and Smad3 in rat wound tissue significantly decreased(P<0.05).Conclusion:Zibai ointment may promote perianal wound healing by regulating the expression of TGF-β1 and Smad3 in rat wound tissue.
