摘要
目的建立黑老虎种子检验方法,为黑老虎种子质量标准的制定提供依据。方法参照《农作物种子检验规程》实施指南及相关文献,以不同试剂、浸种方式及时间组合处理两批黑老虎种子,通过发芽率筛选最适播种前处理方法;考察不同贮藏方式和时间对发芽率的影响;采用正交试验优化黑老虎种子的红四氮唑(TTC)生活力测定参数;并对黑老虎种子进行水分测定和质量测定。结果黑老虎种子播前处理的最优方法为40℃纯水浸种2 h,室温纯水浸种10 h;发芽试验条件为于自然光照下室温土培,播种后第60天计数。经最优前处理方法处理后的两批次黑老虎种子发芽率分别为60%、88%;此外,种子经密封冷藏6周发芽率可达最高,后快速下降。黑老虎种子生活力的最佳条件为种子穿刺后预湿6 h,用0.3%TTC于40℃避光染色24 h。分别采用低恒温烘干法和百粒法测定两批种子的千粒重分别为210.97、218.18 g,含水量分别为13.15%、18.64%。结论建立了黑老虎种子质量检验方法,黑老虎种子采后密封冷藏有助于一定时间内提高黑老虎种子发芽率。
Objective To set up the quality inspection procedures for Kadsura coccinea seeds,and to provide basis for the establishment of seed quality standard of this folk medicinal plant.Methods The inspection procedures for seeds of Kadsura coccinea were studied with reference to the implementation guidelines of Crop Seed Inspection Procedures and literatures.Taking the germination percentage as index,two batches of seeds were investigated by immersing seeds with three combinations of different chemicals,seed-treating methods,and duration to understand the suitable method of presowing treatment.The effects of different storage conditions(at room temperature unsealed,or in fridge with sealed bag for different period)and time on the germination percentage of K.coccinea seeds were also tested.Indexes for viability determination of K.coccinea seeds using of 2,3,5-triphenyl tetrazolium chloride(TTC)were optimized by orthogonal test.The moisture determination and weight determination of K.coccinea seeds were also carried out according to the guidelines.Results The optimal pretreatment method for K.coccinea seeds was to immerse seeds in pure water for 2 hours at 40℃,followed by another 10 hours at room temperature.For germination test,pretreated seeds should be sown on organic soil and kept at room temperature with natural light and the duration of the test is 60 days.The germination rates of the two batches of K.coccinea seeds pretreated under this condition were 60%and 88%,respectively.The seed germination percentage reached the highest after 6 weeks of storing in sealed bag under refrigeration,and then decreased rapidly.The best method for viability determination of K.coccinea seeds was to presoak the seeds for 6 hours after puncture,and stain the seeds with 0.3%TTC solution at 40℃for 24 hours in dark.The moisture determination and weight determination were conducted using a low-constant-temperature oven method and a hundred-grain method,respectively.The thousand grain weights of two batches of seeds were 210.97 g and 218.18 g,and the water contents were 13.15%and 18.64%,respectively.Conclusion The seed quality inspection procedures of K.coccinea were established,and it was found that cold storage of seeds in sealed bag for a certain time could improve the germination percentage of K.coccinea seeds.
