正交试验优化苦豆子复方抗白色念珠菌物质的醇提工艺

摘要 本试验旨在用苦豆子复方醇提物对白色念珠菌的最小抑菌浓度(MIC)为考察指标以优化提取工艺。先采用五因素五水平单因素试验,每一因素再选取3个较低的水平,按照正交表进行五因素三水平的正交试验。结果显示,苦豆子复方抗白色念珠菌物质的最佳提取温度为60℃,料液比为1∶20,提取时间为2.0 h,提取次数为1次,提取溶剂为65%乙醇;苦豆子复方有一定的抗白色念珠菌活性,MIC值为1.25 mg/mL。结果表明,该提取工艺具有操作简便、条件温和等优点,并且较大限度的提取了有效物质。 This study utilized the minimal inhibitory concentration(MIC)of the ethanol extract of anti-Candida albicans Kudouzi compound as an indicator to optimize the extraction process.First used the five-factor five-level single-factor test,and then selected three lower levels for each factor,and performed the five-factor three-level orthogonal test according to the orthogonal table.The results showed that the optimal extraction temperature of anti-C.albicans Kudouzi compound was 60℃,the material-to-liquid ratio was 1∶20,the extraction time was 2.0 h,the number of extractions was 1 times,and the extraction solvent was 65%ethanol.Kudouzi compound demonstrated certain activity against C.albicans,with an MIC value of 1.25 mg/mL.Thus,the optimized extraction process has the advantages of simple operation and mild conditions,and allows extraction of the effective substances to a greater extent.
