响应面法优化双黄连口服液醇沉物乳酸菌发酵工艺

Optimization of liquid fermentation process of Shuanghuanglian oral liquid alcohol sediment with lactic acid bacteria using the response surface technology

摘要 为研究双黄连口服液制剂醇沉物的乳酸菌液态发酵工艺,首先确定双黄连口服液制剂醇沉物添加量,再以活菌数作为指标,通过单因素试验分别研究接种量、发酵时间和发酵温度的影响,并采用响应面法优选最佳发酵条件,最后进行平行放大试验验证。结果:含20%的醇沉物、接种量4%、发酵温度32.7℃、发酵时间26 h时活菌数可达3.03×10^(9)CFU/mL,为最佳发酵条件。平行放大验证试验表明,该发酵工艺可用于制备双黄连口服液醇沉物乳酸菌液态发酵制剂。 To study the liquid fermentation process of the alcohol sediment of Shuanghuanglian oral liquid(SHL·AS)with lactic acid bacteria,the quantity of live bacteria was used as an evaluation indicator.The amount of SHL·AS added was first determined,and then the effects of inoculation amount,time duration,and temperature on the fermentation process were studied through single factor experiments.The response surface technology was used to optimize the fermentation conditions,and parallel experiments were conducted to verify the results.The results showed the optimal fermentation conditions to be 20%SHL·AS with an inoculation amount of 4%,a temperature of 32.7℃,and a duration of 26 hours,under which the quantity of viable bacteria was as high as 3.03×10^(9)CFU/mL.The parallel validation test results confirmed that these conditions were optimal.This fermentation process might be used to produce SHL·AS lactic acid bacteria liquid fermentation preparation.
