摘要
目的:分析过氧化物还原酶3(PRX3)、胎盘生长因子胚胎生长因子(PLGF)、妊娠特异性糖蛋白1(PSG1)在胚胎停育患者绒毛组织中的表达。方法:选取2020年1月-2022年8月本院收治的84例胚胎停育患者作为观察组,60例正常早孕行人工流产者作为对照组;采用PCR及western blot法检测两组绒毛组织中PRX3、PLGF、PSG1的mRNA及蛋白水平,logistics模型分析绒毛组织PRX3、PLGF、PSG1与胚胎停育关系。结果:观察组PRX3及PSG1 mRNA(3.43±0.84、3.31±1.34)和蛋白水平(1.74±0.39、2.39±0.54)均高于对照组(1.54±0.42、2.26±0.43、1.01±0.27、1.83±0.39),PLGF mRNA(0.34±0.05)和蛋白水平(0.51±0.11)均低于对照组(0.42±0.06、0.93±0.24);绒毛组织中高PRX3、PSG1水平和低PLGF水平均与胚胎停育有关(均P<0.05)。结论:相较于正常早孕,胚胎停育患者绒毛组织中PRX3、PSG1明显偏高,PLGF水平明显偏低。
Objective:To analyze the expressions of peroxidase reductase 3(PRX3),placental growth factor(PLGF),and pregnancy-specific glycoprotein 1(PSG1) in the villus tissues of women with embryonic damage,and to study their correlation with the state of embryonic damage of the women.Methods:A total of 84 women with embryonic damage were selected in observation group and 60 women with normal early pregnancy who wanted abortion were selected in control group from January 2020 to August 2022.The levels of the mRNA and the protein of PRX3,PLGF,and PSG1 in the villous tissues of the women in the two groups were detected by PCR and western blot.Logistic model was used to analyze the correlation between the levels of PRX3,PLGF,and PSG1 in the villous tissues of the women and their embryonic damage situation.Results:The mRNA levels of PRX3 and PSG1(3.43±0.84 and 3.31±1.34) and protein levels(1.74±0.39 and 2.39±0.54) of the women in the observation group were significantly higher than those(1.54±0.42,2.26±0.43,1.01±0.27,and 1.83±0.39) of the women in the control group.The levels of PLGF mRNA(0.34±0.05) and protein(0.51±0.11) of the women in the observation group were significantly lower than those(0.42±0.06 and 0.93±0.24) of the women in the control group.The high levels of PRX3 and PSG1 and the low PLGF level in the villus tissues of the women were associated with their embryo damage(all P<0.05).Conclusion:Compared with those in the villus tissues of the women with normal early pregnancy and abortion,the PRX3 and PSG1 levels in the villus tissues of the women with embryo damage are significantly higher,and the PLGF level is significantly lower.
