染色体全自动扫描仪在低比例嵌合体核型分析中的应用

摘要 目的:为了提高低比例嵌合体核型分析的性能,研究染色体全自动扫描仪在该方面的作用,从而发现可改进的部分,以提高低比例嵌合体核型检出率。通过与传统方法比对,探讨染色体全自动扫描仪在低比例嵌合体核型分析中的价值。方法:收集在本院进行诊断、咨询的2000例女性患者作为研究对象,采用数字表法随机分为对照组(n=1000)和研究组(n=1000),对照组采用显微镜手动分析法,研究组采用染色体全自动扫描仪系统分析法,取外周血液淋巴细胞进行培养,常规染色体采用G显带技术核型分析,对照组和研究组均分析计数30个中期分裂相,分析5个核型,出现异常者则加倍核型分析,计数100个核型。结果:通过观察计数,对照组中,检查时出现低比例嵌合体5例,检查低比例嵌合体率为0.5%;研究组中,检查时出现低比例嵌合体20例,检查低比例嵌合体率为2%,查出率显著上升,差异具有统计学意义(P<0.05)。结论:相比传统手动核型分析方法,染色体全自动扫描仪在低比例嵌合体核型分析方面,提高了低比例嵌合体核型的检出率和准确度,给临床提供诊断参考,具有较高的应用价值。 Objective:In order to improve the performance of low-proportion chimera karyotyping,it is necessary to study the role of chromosome automatic scanner in this regard,so as to find points that can be improved to improve the detection rate of low-proportion chimera karyotyping.By comparing with traditional methods,the value of chromosome automatic scanner in low-proportion chimera karyotyping analysis was discussed.Methods:A total of 2000 female patients who came to our hospital for diagnosis and consultation were collected as research objects,and were randomly divided into control group(n=1000)and research group(n=1000)by digital table method,the control group was manually analyzed by microscope,the research group was analyzed by chromosome automatic scanner system,peripheral blood lymphocytes were cultured,conventional chromosomes were karyotyped by G band technology,and 30 metaphase phases were analyzed and counted in the control group and research group,and 5 karyotypes were analyzed.If there is an abnormality,the karyotype analysis is doubled,and 100 karyotypes are counted.Results:Through observation and counting,5 cases of low-proportion chimeras appeared in the control group,and the rate of low-proportion chimeras was 0.5%.In the research group,there were 20 cases of low proportion chimerism during examination,and the low proportion chimerism rate was 2%.The detection rate significantly increased,and the difference was statistically significant(P<0.05).Conclusion:Compared with the traditional manual karyotyping method,the chromosome automatic scanner improves the detection rate and accuracy of low-proportion chimera karyotyping in the analysis of low-proportion chimera karyotyping,which provides a diagnostic reference for clinical practice and has high application value.
