核桃DAHPS基因家族的鉴定及表达分析

摘要 为分析核桃DAHPS基因家族的理化特征和表达模式,阐明该基因家族在核桃中的潜在功能和作用,本研究利用HMM搜索鉴定核桃基因组中的DAHPS基因家族,使用生物信息学分析方法对DAHPS蛋白的理化性质、三维结构和进化关系进行分析,采用qRT-PCR方法对DAHPS基因家族在不同组织中的表达模式进行分析。结果表明:在核桃中共鉴定出6个DAHPS基因家族成员,分布在5条染色体上。编码蛋白对应的氨基酸数为307~540,相对分子质量为33.78~60.04 kDa,理论等电点为6.40~8.57,均含有DAHP_synth_2超家族结构域;除Jr01_17120外,金属离子结合位点Cys-Ser-Ala-His基序在其余5个蛋白中均保守。核桃DAHPS均为亲水性蛋白,具有较好的稳定性。DAHPS基因家族在核桃不同组织中均有表达,不同基因的表达模式不同且与系统进化分析结果相呼应,说明不同DAHPS基因可能存在功能上的分化。研究结果为进一步探究DAHPS基因家族的生物学功能及其在核桃酚类物质代谢中的作用奠定了基础。 HMM search method is used to identify DAHPS gene family in Juglans regia L.genome to analyze the physicochemical characteristics and expression patterns of Juglans regia L.DAHPS gene family as well as elucidate their potential functions and effects.Bioinformatics analysis is used to analyze the physicochemical properties,three-dimensional structure and evolutionary relationship of DAHPS protein.The expression patterns of DAHPS gene family in different tissues are analyzed by qRT PCR.The results show that six DAHPS gene family members are identified and distributed on five chromosomes.The encoded protein has 307~540 amino acids,a relative molecular weight range of 33.78~60.04 kDa and a theoretical isoelectric point range of 6.40~8.57,all containing DAHP_synth_2 superfamily domains.Except for Jr01_17120,the Cys-Ser-Ala-His motif of the metal ion binding site is conserved in the other five proteins.Juglans regia L.DAHPS is a hydrophilic protein with good stability.DAHPS gene family is expressed in different tissues of Juglans regia L.The expression patterns of various genes are different while consistent with the results of phylogenetic analysis,indicating that different DAHPS genes may have functional differentiation.The results lay a foundation for further exploring the biological function of DAHPS gene family and its role in the metabolism of Juglans regia L.phenols.
