摘要
为更全面了解广东省本地EV-A71的流行情况以及分子特征,本研究收集了1994-2021年广东省急性弛缓性麻痹(Acute flaccid paralysis,AFP)监测中分离获得的EV-A71毒株,使用Excel软件统计1994-2021年AFP病例中EV-A71的检出情况,PCR扩增肠道病毒的VP1全长基因片段并测定基因序列,使用BioEdit、iqtree等软件对EVA71进行分型与进化关系分析。本研究共获得119株EV-A71毒株,广东省EV-A71的检出率总体呈先上升后下降的趋势,2010年AFP病例中EV-A71的检出率达到最高,2017年后未从AFP病例样本中检出EV-A71。EV-A71病毒的VP1序列全长891bp,编码297个氨基酸,未见氨基酸插入或缺失。119条EV-A71 VP1序列核苷酸相似性为95.9%(88.4%,100%),氨基酸相似性为99.3%(97.3%,100.0%),均属于EV-A71 C4亚型,其中1994-2001年AFP中分离的EV-A71毒株均为C4b亚型,2004年后流行的EV-A71毒株为C4a亚型。在22位点上,2007年后H取代Q成为22位点上的主要氨基酸,并在2008年出现22R突变;在289位点上,1994年开始出现289T突变,2009年开始,289A(57.1%)取代289T成为优势突变,并于2010年开始出现289V突变。掌握EV-A71随时间迁移的分子遗传进化特征,有利于为EV-A71感染的防控提供科学依据。
To better understand the prevalence and molecular genetic characteristics of EV-A71 in Guangdong Province,China,we collected EV-A71 strains isolated from Acute flaccid paralysis(AFP)surveillance in Guangdong Province,China,from 1994 to 2021.Excel software was used to count the detection rate of EV-A71 in AFP cases from 1994 to 2021.PCR was performed to amplify the VP1 full-length gene of Enterovirus and determine the gene sequence.BioEdit and iqtree were used to analyze the typing and evolutionary relationship of EV-A71.A total of 119 EV-A71 strains were obtained.The detection rate of EV-A71 in Guangdong Province,China,was increasing at first and then decreased.The detection rate of EV-A71 in AFP cases reached the highest in 2010,and was not detected after 2017.The full length of VP1 sequence of EV-A71virus was 891bp,encoding 297 amino acids.No amino acid insertion or deletion was found.The nucleotide similarity of 119 EV-A71 VP1 sequences was 95.9%(88.4%,100%),and the amino acid similarity was 99.3%(97.3%,100.0%).All belong to the C4 subtype of EV-A71.Among them,the EV-A71 strains isolated from AFP from 1994 to 2001 were all C4b subtype.The EV-A71 strains circulating after 2004 belong to subtype C4a.At site 22,H replaced Q as the main amino acid at site 22 after 2007,and 22R mutation appeared in 2008.At locus 289,289T mutation began to appear in 1994,289A(57.1%)replaced 289T as the dominant mutation in 2009,and 289V mutation began to appear in 2010.Understanding the genetic evolution characteristics of EV-A71 over time is conducive to providing scientific basis for the prevention and control of EV-A71 infection.
