摘要
RIG-Ⅰ是维甲酸诱导基因Ⅰ样受体家族(Retinoic-acid-inducible geneⅠ-like receptors,RLRs)的成员之一,在抗病毒固有免疫应答中发挥重要作用。作为细胞质中的一种RNA解旋酶,RIG-Ⅰ可通过其RNA结合结构域与PAMPs结合,识别非自身病毒RNA,触发自身以及下游信号分子的活化,最终诱导IFN-Ⅰ、炎性细胞因子、趋化因子等产生,激活抗病毒免疫应答。本文综述了RIG-Ⅰ在抗病毒识别中的结构特性及在多种RNA病毒与DNA病毒感染中的抗病毒免疫作用或病毒免疫逃逸作用的研究进展,以期为深入研究RIG-Ⅰ抗病毒免疫调控机制及病毒免疫逃逸机制,进而为发现新的抗病毒靶点、研制新型抗病毒药物和疫苗提供理论参考。
RIG-Ⅰis a member of the retinoic-acid-inducible geneⅠ-like receptors(RLRs)family and plays an important role in antiviral innate immune response.As an RNA helicase in the cytoplasm,RIG-Ⅰcan bind to PAMPs through its RNA binding domain,recognizing non-self viral RNA,triggering the activation of itself as well as downstream signaling molecules,and ultimately inducing the production of IFN-Ⅰ,inflammatory cytokines and chemokines to activate antiviral immune responses.This paper reviewed the structural characteristics of RIG-Ⅰin antiviral recognition,and research advances of RIG-Ⅰin antiviral immunity and viral immune evasion in a variety of RNA viruses and DNA viruses infection,aiming to provide theoretical references for the discovery of new antiviral targets and the development of novel antiviral drugs and vaccines based on the mechanisms of antiviral immune regulation and viral immune evasion of RIG-Ⅰ.
