灵芝酸类成分抑制前列腺癌生长及其机制研究 被引量:1

Study on the Inhibition of Ganoderma lucidum Acids on the Growth of Prostate Cancer and its Mechanism

摘要 研究灵芝酸类成分对前列腺癌LNCaP细胞生长的影响,并探讨其相关机制。配置含不同浓度灵芝酸C1、F、G、H、I、K、L、M、N的无色完全培养基,以氟他胺为阳性对照;AlamarBlue?试剂法测定其对LNCaP细胞生长的影响;流式细胞仪检测其对LNCaP细胞周期的影响;荧光定量PCR和蛋白免疫印记试验检测caspase-3、caspase-8、bcl-2、bax基因及相关蛋白的表达情况。结果显示:1.灵芝酸类成分对前列腺癌细胞增殖均具有抑制作用,并且呈浓度依赖性,其中灵芝酸C1、M、N抑制效果明显,低浓度其抑制率与阳性对照相近,而高浓度灵芝酸M、N抑制率高于阳性对照;2.灵芝酸类成分均可以将细胞周期阻滞在有丝分裂间期的G1期和G0期,抑制了癌细胞的进一步增殖,其阻滞能力随给药浓度增加而升高,灵芝酸C1、F、G、H、L、M、N在低浓度时都具有一定抑制细胞分裂的效果,其中灵芝酸C1、F、G、H、L、M、N高浓度与阳性对照相当甚至更高;3.不同浓度的灵芝酸类成分均能上调caspase-3、caspase-8、bax促凋亡基因和下调bcl-2抑凋亡基因的表达,并且都呈现浓度依赖性。灵芝酸类成分尤以灵芝酸C1、M、N能够促进前列腺癌LNCaP细胞的凋亡,因其能阻滞细胞分裂间期处于G1期和G0,同时上调caspase-3、caspase-8、bax促凋亡基因和下调bcl-2抑凋亡基因的表达。 To investigate the effects of Ganoderma lucidum acids on the growth of prostate cancer LNCaP cells and explore the related mechanisms,colorless complete cture medium containing different concentrations of Ganoderma lucidum acid C1,F,G,H,I,K,L,M,N was prepared,with flutamide as the positive control.The growth of LNCaP cells was determined by AlamarBlue®reagent method;The effect of LNCaP on cell cycle was detected by flow cytometry;The expression of caspase⁃3,caspase⁃8,bcl⁃2,bax genes and related proteins were detected by fluorescence quantitative PCR and Western blot.The result showed:1.Ganoderma lucidum acid has inhibi⁃tory effect on the proliferation of prostate cancer cells in a concentration⁃dependent manner;Among them,the inhibitory effect of Ganoderma lucidum acid C1,M and N is obvious,the inhibitory rate of low concentration of Ganoderma lucidum acid M and N is close to that of positive control,while the inhibitory rate of high concentration of Ganoderma lucidum acid M and N is higher than that of positive control.2.Ganoderma lucidum acid can block the cell cycle in G1 and G0 phases of mitosis,inhibit the further prolifera⁃tion of cancer cells,and its blocking ability increases with the increase of drug concentration.Ganoderma lucidum acid C1,F,G,H,L,M,N has certain effect of inhibiting cell division at low concentration,among which the high concentration of Ganoderma lucidum acid C1,F,G,H,L,M,N is equivalent to or even higher than the positive control.3.Ganoderma lucidum acids at different concentra⁃tions can up⁃regulate the expression of caspase⁃3,caspase⁃8,bax pro⁃apoptotic genes and down⁃regate the expression of bcl⁃2 anti⁃apop⁃totic genes,and all of them are concentration⁃dependent.Ganoderic acids,especially Ganoderic acid C1,M and N,can promote the apoptosis of LNCaP cells,because they can block the G1 and G0 interphase of cell division,caspase⁃3,caspase⁃8 and bax were up⁃re⁃gated and bcl⁃2 was down⁃regated.
