黄芪甲苷对高糖受损间充质干细胞生物学功能和分泌HGF的影响

摘要 目的:探讨黄芪甲苷(astragaloside IV,AS-IV)对高糖受损间充质干细胞(mesenchymal stem cells,MSCs)生物学功能和分泌肝细胞生长因子(hepatocyte growth factor,HGF)的影响。方法:无菌条件下取足月健康新生儿胎盘脐带血,经密度梯度离心分离出人脐带血间充质干细胞(human umbilical cord blood mesenchymal stem cells,hUCBMSCs),分别进行成骨、成软骨和成脂诱导分化鉴定。将鉴定成功的hUCBMSCs用30 mmo1/L葡萄糖预处理120 h后,用不同浓度梯度AS-IV(0、50、100、200、300、400 mg/L)进行干预,确定AS-IV促进高糖受损hUCBMSCs增殖的最适浓度。另将高糖受损hUCBMSCs随机分为实验组和模型组,实验组用最适浓度AS-IV处理,模型组用等体积PBS液处理,同时设置正常组,即等量PBS液处理正常hUCBMSCs。通过CCK-8细胞增殖试验、黏附能力测定试验、细胞划痕试验及ELISA试验检测最适浓度AS-IV对hUCBMSCs增殖、黏附、迁移及分泌HGF功能的影响。结果:AS-IV改善受损hUCBMSCs增殖功能的最适浓度为300 mg/L;模型组高糖受损hUCBMSCs的增殖、黏附、迁移及分泌HGF功能低于正常组(P<0.05);实验组高糖受损hUCBMSCs的增殖、黏附、迁移及分泌HGF功能明显改善,与模型组比较差异具有统计学意义(P<0.05)。结论:黄芪甲苷可促进高糖受损间充质干细胞增殖,改善其分泌功能。 Objective:To investigate the effects of AS-IV on the biological functions and secretion of HGF in high glucose-damaged MSCs.Methods:Human umbilical cord blood mesenchymal stem cells(hUCBMSCs)were isolated by density gradient centrifugation under aseptic conditions from placental cord blood of healthy fullterm newborns and identified for osteogenic,chondrogenic and lipogenic induction of differentiation respectively.After successfully identified hUCBMSCs pretreated with 30 mmo1/L glucose for 120 h and then intervened with different gradients of AS-IV(0,50,100,200,300 and 400 mg/L),the optimal concentration of AS-IV was determined to promote proliferation of high glucose-impaired hUCBMSCs.In addition,high glucose-impaired hUCBMSCs were randomized into the experiment group and the model group,the experimental group was treated with the optimal concentration of AS-IV,and the model group was dealt with an equal volume of PBS solution,at the same time,the normal group was set,namely,processing normal hUCBMSCs with equivalent volume of PBS solution.The effect of the optimal concentration of AS-IV on the proliferation,adhesion,migration and HGF secretion of hUCBMSCs was measured by CCK-8 cell proliferation assay,adhesion assay,cell scoring assay and ELISA.Results:The optimal concentration of AS-IV which could improve the proliferation of damaged hUCBMSCs was 300 mg/L;the proliferation,adhesion,migration and HGF secretion functions of hUCBMSCs damaged by high glucose in the model group were lower than these of the normal group(P<0.05);the proliferation,adhesion,migration and HGF secretion functions of hUCBMSCs damaged by high glucose in the experiment group were significantly improved,and the difference was statistically significant(P<0.05).Conclusion:AS-IV can protect high glucose-damaged MSCs and improve their secretory function.
