摘要
空间多组学技术包括空间转录组学、空间蛋白质组学和空间代谢组学等技术。相较于传统测序和单细胞组学,空间多组学保留了重要的分子空间位置,多维度揭示分子间相互作用及功能状态。在肿瘤学领域,空间多组学技术能将肿瘤细胞及其间质二维结构分辨至细胞以及亚细胞水平。空间多组学技术将推动抗肿瘤药物精准选择、给药方式优化、治疗反应监测以及耐药机制探索等多个研究方向,辅助肿瘤精准诊疗。本文概述了空间多组学技术的发展历史,对空间多组学技术进行了分类,总结了各类方法的代表性技术手段及检测原理。列举了空间多组学技术在消化系统肿瘤中的研究及应用,并对其未来的发展进行了展望。
Spatial omics techniques include spatial transcriptomics,spatial proteomics,and spatial metabolome.Compared with traditional sequencing and single-cell omics,spatial omics preserves the spatial location and reveals the interaction between molecules.In the field of oncology,spatial omics techniques can deconstruct the two-dimensional molecular structure of tumors to the cellular or even subcellular level.Spatial omics technology will promote the precise selection of anti-tumor drugs,the optimization of administration methods,the monitoring of treatment response.It can also provide support for the exploration of drug resistance mechanisms and the precise diagnosis and treatment of tumors.This paper summarized the development of spatial omics technology,classified spatial omics technology,and summarized the representative technical methods and detection principles of various methods.We also reviewed research and application of spatial omics technology in gastrointestinal tumors and evaluated the future development.
