摘要
采用亲水作用色谱法(HILIC)测定达原饮中槟榔生物碱的含量,4种槟榔生物碱分离度良好,各组分在浓度范围内线性关系良好,加标回收率(n=9)98.77%~102.87%,RSD值均小于2.0%。经过样品测试,达原饮药液中去甲槟榔碱含量为0.0209 mg•mL^(-1),槟榔碱含量为0.0679 mg•mL^(-1),去甲槟榔次碱含量为0.0545 mg•mL^(-1),槟榔次碱含量为0.0269 mg•mL^(-1)。该检测方法简便、准确、灵敏、重复性好,可用于测定达原饮中槟榔生物碱的含量。
A method for the determination of areca alkaloids in Dayuanyin was established by Hydrophilic Interaction High Performance Liquid Chromatography(HILIC).The chromatographic conditions were as follows:Agela Venusil HILIC propyl amide bonded silica gel column(4.6 mm×250 mm,5µm,Tianjin Agela Technology Co.,LTD.);Mobile phase:acetonitrile:0.1%triethylamine(pH adjusted to 3.5 by phosphoric acid)=70:30;Detection wavelength:210 nm;Column temperature:30℃;Flow rate:1.0 mL•min^(-1).The four areca alkaloids were well separated,and the linear relationship of each component was good in the concentration range.The standard recoveries(n=9)ranged from 98.77%to 102.87%,and RSD were all lower than 2.0%.The sample test showed that the contents of guvacoline,arecoline,guvacine hydrochloride and arecaine in Dayuan decoction were 0.0209 mg•mL^(-1),0.0679 mg•mL^(-1),0.0545 mg•mL^(-1) and 0.0269 mg•mL^(-1).The method is simple,accurate,sensitive and reproducible,and can be used for the determination of areca alkaloids in Dayuanyin.
