摘要
采用单因素-响应面法对超声辅助乙醇提取总黄酮的条件进行优化,在最佳条件(乙醇浓度55%、料液比1∶30 g/mL、提取温度43℃)下,蝴蝶花总黄酮实际提取率为42.58%,该抗氧化能力研究发现蝴蝶花总黄酮对·OH与NO_(2)^(-)有一定的清除能力.为提高学生分析问题、解决问题的能力,将蝴蝶花总黄酮的提取、含量测定及抗氧化性能研究设计为综合实验,通过该实验,可提高学生文献检索、方案设计、数据处理分析等系统性实验技能,培养其团队协作和探索创新的意识.
The single factor-response surface methodology was used to optimize the conditions for ultrasonic assisted ethanol extraction of total flavonoids from Iris japonica Thunb.Under the optimal conditions(ethanol concentration 55%,solid-liquid ratio 1:30 g/mL,extraction temperature 43 ℃),the actual extraction rate of total flavonoids was 42.58%.It was found that the total flavonoids of Iris jqponica Thunb had a certain scavenging ability to OH and NO_(2)^(-).In order to improve students'ability to analyze and solve problems,the comprehensive experiment was designed on the extraction,content determination and antioxidant performance of total flavonoids from Iris Japonica Thunb.By this comprehensive experiment,students'systematic experimental skills in document retrieval,scheme design,data processing and analysis were improved,their awareness of teamwork and exploration and innovation were cultivated.
