摘要
[目的]筛选与茶树炭疽病存在拮抗作用的土壤细菌,制备高效拮抗菌剂。[方法]采用平板对峙法筛选出对茶树胶孢炭疽菌具有较强拮抗作用的菌株,通过形态学观察、生理生化试验和16S rDNA分子水平进行种属鉴定,采用响应面法优化液体培养条件。[结果]筛选出的菌株Y-8为萎缩芽孢杆菌(Bacillus atrophaeus)。该菌株对病原菌菌丝生长抑制率为53.2%,具有产纤维素酶、淀粉酶和蛋白酶的能力。生长曲线测定发现,Y-8菌株对数生长期为接种后2~18 h。响应面法优化后的培养条件为:麸皮添加量4.5%(w/v)、(NH_(4))_(2)SO_(4)添加量1.0%(w/v)、接种量5.0%(v/v)。[结论]Y-8菌株繁殖迅速、易培养,可开发为高效且低成本的茶树炭疽病生防菌剂。
[Purpose]Antagonistic soil bacteria was screened to prepare microbial agent against Gloeosporium theae-sinensis Miyake.[Method]Bacterial strain with antagonistic effect on G.theae-sinensis was screened by plate standoff method,and was further identified through morphological observation,physiological and biochemical tests and 16S rDNA phylogenetic analysis.Then response surface methodology was used to optimize the optimal culture conditions.[Result]One Bacillus atrophaeus strain named Y-8 was screened,with an inhibitory effect of 53.2%against Colletotrichum gloeosporioides,and it was capable of producing cellulase,amylase and protease.Its logarithmic growth phase took place 2-18 h after inoculation.The optimal culture conditions inferred from response surface methodology were to use 4.5%(w/v)wheat bran and 1.0%(w/v)(NH_(4))_(2)SO_(4)at an inoculum dosage of 5.0%(v/v).[Conclusion]The study laid an foundation for the development of strain Y-8 as an biocontrol agent against G.theae-sinensis.
