姜黄素对血管性痴呆大鼠肠道菌群代谢及记忆功能障碍改善的作用机制分析 被引量:1

Mechanism of Curcumin on Improvement of Intestinal Flora Metabolism and Memory Dysfunction in Rats with Vascular Dementia

摘要 目的分析姜黄素对血管性痴呆大鼠肠道菌群代谢及记忆功能障碍改善的作用机制。方法使用配对比较法,将45只SD大鼠分别纳入试验组、模型组、对照组各15只。试验组、模型组均建立血管性痴呆大鼠模型,对照组行假手术处理。成功建模后给予试验组300 mg/kg姜黄素灌胃,每日1次,持续4周;模型组、对照组同期给予同等剂量生理盐水灌胃。收集各组大鼠处理4周后粪便样本,检测其肠道菌群结构;运用Morris水迷宫实验评价各组大鼠处理4周后认知和记忆功能变化。结果试验组d1~d5逃避潜伏期均低于模型组,穿越平台次数、游动速度高于模型组;对照组d1~d5逃避潜伏期均低于试验组,穿越平台次数、游动速度高于试验组(P<0.05)。3组大鼠肠道菌群检测共检出OTUs 4917个,3组间共有OTUs为525个。与对照组比较,模型组Shannon指数、Chao1指数均升高,试验组Shannon指数、Chao1指数略低于模型组,组间比较未见统计学意义(P>0.05)。主成分分析显示,模型组肠道菌群结构与对照组、试验组存在明显统计学意义(P=0.037)。模型组瘤胃菌科Ruminococcaceae、棒状杆菌目Corynebacteriales、棒状杆菌属Corynebacterium_1和Kineothrix丰度上调,试验组Robinsoniella丰度上调,差异有统计学意义。试验组iNOS蛋白含量低于模型组,高于对照组;试验组、对照组氧自由基、羟自由基水平均低于模型组(P<0.05)。结论血管性痴呆大鼠肠道菌群代谢存在显著变化,姜黄素可能通过调节肠道菌群结构、减少iNOS及自由基过量表达,促进大鼠记忆功能障碍的改善。 Objective To analyze the mechanism of curcumin improving intestinal flora metabolism and memory dysfunction in rats with vascular dementia.Methods Forty-five SD rats were divided into experimental group,model group and control group,with 15 rats in each group.The experimental group and the model group established the vascular dementia rat models,while the control group received sham operation.After successful modeling,the experimental group was given curcumin(300 mg/kg)by in-tragastric administration once a day for 4 weeks.The model group and the control group were given the same dose of normal sa-line at the same time.The fecal samples of rats in each group were collected after 4 weeks of treatment,and the intestinal flora structure was detected.Morris water maze test was used to evaluate the changes of cognitive and memory functions of rats in each group after 4 weeks of treatment.Results The escape latency of d1-d5 in the experimental group was lower than that in the mod-el group,and the times of crossing the platform and swimming speed were more than those in the model group.The escape latency of d1-d5 in the control group was lower than that in the experimental group,and the times of crossing the platform and swimming speed were more than those in the experimental group(P<0.05).A total of 4917 OTUs were detected in the intestinal flora of rats in the three groups,and 525 among the three groups.Compared with those of the control group,Shannon index and Chao1 in-dex of the model group were all increased,while Shannon index and Chao1 index of the experimental group were slightly lower than those of the model group,and there was no statistical difference between the two groups(P>0.05).Principal component a-nalysis showed that the intestinal flora structure of the model group was significantly different from that of the control group and the experimental group(P=0.037).The abundance of rumen bacteria Ruminococcus,Corynebacterium,Corynebacterium_1 and Kineothrix in the model group was up-regulated,while that of Robinsoniella in the experimental group was up-regulated,with significant difference.The content of iNOS protein in the experimental group was lower than that in the model group,but higher than that in the control group.The levels of oxygen free radical and hydroxyl free radical in the experimental group and the con-trol group were lower than those in the model group(P<0.05).Conclusion There are significant changes in intestinal flora me-tabolism in rats with vascular dementia.Curcumin may promote the improvement of memory dysfunction in rats by regulating in-testinal flora structure and reducing the excessive expressions of iNOS and free radicals.
