貂熊线粒体基因组全序列分析及其系统进化

摘要 为了更好地了解貂熊(Gulo gulo)的起源、进化等生物学信息,采用PCR方法克隆貂熊的线粒体基因组全序列,利用测序技术等常规分子生物学手段对其进行分析。结果显示:貂熊基因组序列全长为16575 bp;同其他哺乳动物相同,含13种蛋白质、22种转运RNA(tRNA)、2种核糖体RNA(rRNA)和1个非编码控制区(D-loop)。4种碱基组成分别为:A=32.18%、G=14.26%、T=26.75%、C=26.81%;AT含量(58.93%)>CG含量(41.07%),存在显著的AT富集。基于基因组全序列数据,以棕熊(Ursus arctos)为外群,采用邻接法和最大似然法构建鼬科(Mustelidae)22种动物的进化树,结果显示貂熊同貂属(Martes)动物关系较近,貂熊同7种貂属动物聚成一支后,再同猪獾(Arctonyx collaris)和狗獾(Meles meles)聚成一支组成并系群,结果证实貂熊同渔貂(Martes pennanti)和黄喉貂(M.flavigula)的亲缘关系最近。 In order to better explore the biological information such as the origin and evolution of wolverine(Gulo gulo),PCR was used to obtain the complete sequence of wolverine mitochondrial genome,conventional molecular biological methods such as sequencing technology were used to analyze.Results showed that the total genome sequence of wolverine is 16,575 bp,which is comparable to other mammals and includes 13 proteins,22 tRNAs,two rRNAs and one control region.The base composition is:A=32.18%,G=14.26%,T=26.75%,and C=26.81%.AT content(58.93%) is greater than CG content(41.07%),and the difference is significant.Based on the full sequence data of the genome,the evolutionary tree of 22 Mustelidae species was constructed using neighbor-joining(NJ) and maximum likelihood(ML) methods with brown bear(Ursus arctos) as the outer group.The findings showed that the relationship between wolverine and marten was relatively close.After the wolverine and seven species of marten gathered into a branch,it formed a parallel group with hog badger(Arctonyx collaris) and European badger(Meles meles).The results also confirmed that wolverine had the closest genetic relationship with fisher marten(Martes pennanti) and yellow-throated marten(M.flavigula).
