基于细菌毒素的肿瘤基因靶向治疗的研究进展

摘要 理想的肿瘤基因靶向治疗方案须具备高效性、特异性、安全性、可调控性等特征。由于细菌毒素毒性强,在肿瘤治疗方面具有高效性,因此,利用细菌毒素与抗体或细胞因子等合成的免疫毒素在临床治疗上得到应用。然而免疫毒素具有免疫原性、非特异性细胞毒性等缺点,于是目前正在探索开发基于细菌毒素的基因靶向治疗。细菌毒素基因靶向治疗可以通过适宜的载体递送系统将毒素基因转入肿瘤细胞,利用肿瘤特异性启动子驱动毒素基因在肿瘤细胞内大量表达,实现毒素基因靶向肿瘤细胞的特异性及效应性,达到抑制肿瘤细胞生长的目的。本文就此过程中涉及的细菌毒素、载体递送系统、肿瘤特异性启动子等方面进行综述,为细菌毒素在肿瘤基因靶向治疗中的应用提供更多理论基础。 The ideal tumor gene-targeted therapy scheme should have the characteristics of high efficiency,specificity,safety and regulation.Because bacterial toxins are highly toxic and highly effective in tumor treatment,immunotoxins synthesized by bacterial toxins and antibodies or cytokines have been used in clinical treatment.However,immunotoxins have disadvantages such as immunogenicity and non-specific cytotoxicity,so the development of gene-targeted therapy based on bacterial toxins is being explored.Bacterial toxin gene targeting therapy is to transfer toxin gene into tumor cells through appropriate carrier delivery system and use tumor-specific promoter to drive the toxin gene expression in large quantities in tumor cells,so as to realize the toxin gene targeted tumor cell specificity and effectiveness,achieve the purpose of inhibiting tumor cell growth.This article reviews the bacterial toxins,carrier delivery systems,and tumor-specific promoters involved in this process,so as to provide more theoretical basis for the application of bacterial toxins in tumor gene targeted therapy.
