摘要
目的探讨NF-κB/P65分别在食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)细胞和肿瘤浸润淋巴细胞(tumor-infiltrating lymphocytes,TILs)中的表达对患者预后的意义。方法回顾性收集80例ESCC的临床病理资料,免疫组化染色法检测NF-κB/P65分别在ESCC细胞和TILs中的表达,分析癌症基因组图谱(the cancer genome atlas,TCGA)数据库中NF-κB/P65在ESCC中的功能。结果ESCC细胞中NF-κB/P65表达阳性率高于正常鳞状上皮,非角化型鳞癌细胞中高于角化型鳞癌,与有丝分裂数呈正相关,癌细胞中NF-κB/P65阳性表达率高的患者预后差(P<0.05)。ESCC中TILs在临床1期高于临床2、3期,与癌灶大小呈负相关(P<0.05)。ESCC中NF-κB/P65在癌周和癌内TILs中的表达与TILs的量呈正相关,癌周TILs中NF-κB/P65表达阳性率高于癌内TILs,NF-κB/P65阳性表达率在原位癌癌周TILs中明显高于浸润性癌周TILs,癌内和癌周TILs中NF-κB/P65阳性表达率高的患者预后较好(P<0.05)。生物信息学分析显示NF-κB/P65与DNA修复、上皮间质转化、基质形成、炎性反应和IL-10抗炎信号通路相关因子呈正相关,与T淋巴细胞呈负相关(P均<0.05)。结论NF-κB/P65在ESCC细胞中的高表达可能促进癌组织的发生进展,在ESCC中,TILs的高表达对ESCC的进展可能具有抑制作用,NF-κB/P65可能分别通过不同的信号通路和分子,影响其在癌细胞和TILs中的不同作用。
Objective To investigate the implications for patient outcomes between the expression of NF-κB/P65 both in esophageal squamous cell carcinoma(ESCC)and tumor-infiltrating lymphocytes(TILS).Methods The clinicopathological data of 80 ESCC cases were retrospectively collected.The expression of NF-κB/P65 in ESCC cancer cells and TILs was detected by immunohistochemical staining,and the function of NF-κB/P65 in ESCC was analyzed in the TCGA database.Results The positive rate of NF-κB/P65 expression in ESCC cancer cells was significantly higher than that in normal squamous cell,and that in non-keratinized squamous cell was obviously higher than that in keratinized squamous cell,as well it was significantly positively correlated with mitotic number,the NF-κB/P65 higher positive expression group in cancer cells had poor prognosis(P<0.05).In TILs of ESCC was significantly higher in clinical stage 1 than that in stage 2 and 3,and it had a significant negative correlation with cancer focus size(P<0.05).The expression of NF-κB/P65 in TILs around and within ESCC was positively correlated with the amount of TILs(P<0.05).The positive expression of NF-κB/P65 in TILs around ESCC was significantly higher than that within ESCC.The positive NF-κB/P65 expression in TILs around ESCC in carcinoma was significantly higher than that in invasive ESCC.Higher NF-κB/P65 positive expression in TILs both around and within ESCC had better prognosis.Biogenic analysis showed that NF-κB/P65 was positively correlated with DNA repair,epithelia-mesenchymal transformation,stroma formation,inflammatory response,and IL-10 anti-inflammatory signaling pathway,but negatively correlated with T lymphocytes(P all<0.05).Conclusion The higher expression of NF-κB/P65 in ESCC cancer cells may promote the development and progression of ESCC,but the higher expression of TILs in ESCC may have an inhibitory effect on the progression of ESCC.NF-κB/P65 may have different effects on cancer cells and TILs through different signaling pathways and molecules respectively.
