摘要
目的观察坤安煎剂对去卵巢大鼠肠道菌群及脂代谢的影响,探讨其疗效机制,为临床治疗提供思路。方法选用SPF级8周龄雌性SD大鼠44只,体质量(200±20)g。去卵巢后将大鼠按随机数表法分4组,空白组和模型组大鼠给予生理盐水,中药组大鼠给予坤安煎剂,调脂药组大鼠给予阿托伐他汀钙片,去卵巢1周后,灌服药物,每天1次,连续10周,检测大鼠血清FSH、E2和血脂水平,每组收集6只大鼠粪便样本并提取粪便DNA,通过16S rDNA高通量测序技术进行检测并分析结果。结果去卵巢后大鼠体质量以及FSH、TC和TG水平显著增加,E2水平显著下降,中药坤安煎剂能显著降低去卵巢大鼠体质量以及FSH、TC、TG水平,增加血清E2水平,且在降低FSH水平、升高E2水平上优于调脂药组(均P<0.05)。基于16S rDNA测序对大鼠肠道菌群进行分析,韦恩图显示两组间物种组成差异较大。Alpha多样性指数显示,中药组大鼠肠道菌群的多样性显著高于调脂药组。Beta多样性分析显示,4组大鼠肠道菌群结构具有显著差异。属水平物种丰度显示,中药组大鼠肠道菌群Prevotella、Bacteroides、Parabacteroides的丰度高于调脂药组(均P<0.05)。进一步对属水平物种相关性高的肠道菌群和血脂水平进行相关性分析,发现肠道Prevotella、Bacteroides、Parabacteroides与TG水平呈负相关。结论坤安煎剂能显著降低去卵巢大鼠体质量以及FSH、TC、TG水平,增加血清E2水平,且在降低FSH水平、升高E2水平上优于调脂药组。去卵巢后大鼠肠道菌群紊乱,坤安煎剂干预可以显著改善肠道菌群紊乱,并且可通过调节Prevotella、Bacteroides、Parabacteroides的丰度来改善血脂水平。
Objective To observe the effect of Kun'an decoction on intestinal microbiota and lipid metabolism in ovariectomized rats,discuss the therapeutic mechanism,and provide ideas for better clinical treatment of this disease.Methods Forty-four 8-week-old SPF female SD rats weighing(200±20)g were selected.After ovariectomy,the rats were randomly divided into 4 groups;the blank group and model group were given normal saline,traditional Chinese medicine(TCM)group was given Kun′an decoction,and lipid-regulating drug group was given atorvastatin calcium tablets.One week after ovariectomy,the drugs were given once a day for 10 weeks.Serum levels of FSH,E2 and blood lipid of rats were detected.Feces of 6 rats in each group were collected and fecal DNA was extracted.The samples were detected and results analyzed with 16S rDNA high-throughput sequencing technology.Results After ovariectomy,the rats'body weight,FSH,TC and TG increased significantly,while E2 decreased significantly.Kun'an decoction significantly reduced the body weight,FSH,TC and TG,while increased serum E2 level in ovariectomized rats,and was superior to lipid-regulating drugs in reducing FSH and increasing E2(all P<0.05).Based on 16S rDNA sequencing,the intestinal microbiota of rats was analyzed,and Wayne diagram showed that there was a great difference in species composition between the two groups.Alpha diversity index showed that the community diversity of TCM group was significantly higher than that of lipid-regulating drug group.Beta diversity analysis showed that there were significant differences in microbiota structure among the four groups.The abundance of Prevotella,Bacteroides and Parabacteroides in TCM group was higher than that in lipid-regulating drug group(all P<0.05).Further analysis of the correlation between highly species-related intestinal microbiota and blood lipid levels at genus level showed that intestinal Prevotella,Bacteroides and Parabacteroides were negatively correlated with TG level.Conclusion Kun'an decoction can significantly reduce body weight,FSH,TC and TG in ovariectomized rats,and increase serum E2 levels;it is superior to lipid-regulating drugs in reducing FSH and increasing E2 levels.After ovariectomy,the intestinal microbiota of rats was disordered.Kun'an decoction intervention can significantly improve intestinal microbiota disorder,and improve blood lipid level by regulating the abundance of Prevotella,Bacteroides and Parabacteroides.
