野生乌饭树叶片愈伤组织的诱导及增殖 被引量:1

摘要 为建立乌饭树(Vaccinium bracteatum Thunb.)叶片组织培养体系,以野生乌饭树(L-3 D-6)的嫩叶为外植体,探究不同消毒方式、不同浓度激素对乌饭树叶片组织培养增殖的影响。结果表明:采用75%乙醇消毒30 s+0.1%HgCl2+吐温20溶液消毒3 min,外植体消毒效果最好,污染率低至3.33%,褐化率为0;乌饭树嫩叶最佳愈伤组织诱导培养基为:WPM+2.50 mg/L ZT+0.75 mg/L NAA,此培养基培养下,愈伤组织形成时间最早,最终诱导率高达98.15%;乌饭树愈伤组织增殖培养基为WPM+0.30 mg/L ZT+0.50 mg/L NAA,增殖效果最佳,增殖系数达7.93,愈伤组织质地疏松无褐化。 The young leaves of Vaccinium bracteatum Thunb.were used as explants to explore the effects of different disinfection methods and different concentr ations of hormones on the proliferation of leaf fragments in tissue culture.The results showed that when the explants were disinfected with 75%alcohol for 30 s+0.1%HgCl2+Tween 20 solution for 3 min,the disinfection effect was the best,the contamination rate was only 3.33%,and the browning rate was 0.The optimal medium for callus induction was WPM+2.50 mg/L ZT+0.75 mg/L NAA.Under this medium,the callus formation time was the earliest and the final induction rate reached 98.15%.The medium of WPM+0.30 mg/L ZT+0.50 mg/L NAA was the best medium for callus proliferation,and the propagation coefficient reached 7.93.The texture of callus was loose and no browning was found.
