抗无乳链球菌天然产物的筛选

摘要 基于MedChemExpress天然产物库筛选对无乳链球菌有抗菌效果的天然产物,采用成药性预测工具Swisse-ADME分析其成药性,并测定抗菌天然产物对人源、奶牛源、鱼源等不同来源菌株的最小抑菌浓度、最小杀菌浓度和最小生物被膜清除浓度。结果表明:从2443个天然产物中共鉴定出104个对无乳链球菌有显著抗菌效果的天然产物,其中15个成药性前景较好的天然产物,其最小抑菌浓度介于4~256μg·mL^(-1)之间,藤黄酸、血根碱、白屈菜红碱、冬凌草甲素和积雪草酸等5个天然产物对无乳链球菌的最小杀菌浓度介于4~64μg·mL^(-1)之间,最小生物被膜清除浓度介于32~128μg·mL^(-1)之间,具有开发成新型无乳链球菌抗菌药物的潜力。 Based on the MedChemExpress natural products library, the natural products with antibacterial effect against Streptococcus agalactiae were screened. Drug ability of these natural products was analyzed with the druggability prediction tool Swisse-ADME, and the antibacterial natural products were tested for strains from different sources such as human, dairy, and fish for testing minimum inhibitory concentration(MIC), minimum bactericidal concentration(MBC), and minimum biofilm eradication concentration(MBEC). The results showed that 104 natural products with significant antibacterial effects on S.agalactiae were identified from 2 443 natural products, and the MICs of 15 natural products with good druggability were between 4 to 256 μg·mL^(-1). The MBCs of 5 natural products including gambogic acid, sanguinarine, chelerythrine, oridonin and asiatic acid against S.agalactiae ranged from 4 to 64 μg·mL^(-1), and the MBECs were between 32-128 μg·mL^(-1), which had the potential to be developed as new antibacterial drugs for S.agalactiae.
