疏肝利胆颗粒提取工艺优选及药效学实验研究

摘要 目的 优选疏肝利胆颗粒的提取工艺条件,并对其肝保护作用进行评价,为该制剂的开发提供科学实验依据。方法 采取正交试验法,以干膏得率、大黄素和大黄酚的转移率为指标,考察加水量、提取时间、提取次数3个因素,筛选疏肝利胆颗粒最优水提工艺;同时考察不同剂量水提物对α-异硫氰酸萘酚(alpha-naphthylisothiocyanate,ANIT)诱导的小鼠胆汁淤积的治疗作用,明确水提物药效。结果 疏肝利胆颗粒最佳水提工艺为加10倍量水,提取3次,每次1.5 h;与模型组比较,小鼠给予水提物7 d后,血清生化指标和肝组织炎症因子mRNA水平明显降低,水提物可明显缓解肝组织病理学损伤程度。结论 优选的提取工艺稳定可行,药效学研究结果提示消炎利胆方水提物对ANIT诱导的胆汁淤积型小鼠肝损伤具有一定的保护作用。 Objective To optimize the extraction process conditions of Shugan Lidan Granules,and preliminary evaluation of its liver protection effect,to provide a basis for the development of the preparation.Methods Orthogonal test method was used to evaluate the three factors of water addition,extraction time and extraction times based on the yield of dry paste,the transfer rate of emodin and chrysophanol,and the best water extraction process of Shugan Lidan Granules.Investigate the therapeutic effects of different doses of water extracts on alpha-naphthylisothiocyanate-induced cholestasis in mice,and clarify the efficacy of water extracts.Results The best water extraction process was to add 10 times the amount of water and extract three times for 1.5 h each time.Compared with the model group,7 days after administration,serum biochemical indicators and liver tissue inflammatory factor mRNA levels were significantly reduced,and the degree of liver tissue pathological damage was significantly reduced.Conclusion The optimal extraction process was stable and feasible,and the results of pharmacodynamic studies suggest that the water extract of Xiaoyan Lidan Recipe showed certain protective effects on the liver injury of cholestatic induced by ANIT in mice.
