脂多糖诱导大鼠胸主动脉内皮源性外泌体的miRNA表达

摘要 目的 研究脓毒血症早期血管内皮源性外泌体所携带的miRNA的表达情况。方法 体外培养SD大鼠胸主动脉内皮细胞,给予细菌脂多糖诱导处理后,收集的培养基经过差速超速离心后获得外泌体。通过高通量测序检测内皮源性外泌体所携带的小RNA表达,和静态培养的内皮细胞源性外泌体进行比较,找出差异性表达的miRNA。结果 本研究成功分离了来源于血管内皮细胞的外泌体,在透射电镜下观察到直径为40 nm~100 nm的盘状囊泡。对外泌体携带的miRNA进行测序,发现在细菌脂多糖的作用下内皮细胞通过外泌体释放大量差异表达的miRNA。结论 脓毒血症早期血管内皮源性外泌体携带的miRNA表达谱发生改变,这些差异表达的miRNA可能在脓毒血症早期心血管的病理生理学上发挥着重要作用。 Objective To study the expression of miRNAs carried by vascular endothelial-derived exosomes in the early stage of sepsis.Methods SD rat thoracic aortic endothelial cells were cultured in vitro,treated with bacterial lipopolysaccharide,and the collected medium was subjected to differential ultracentrifugation to obtain exosomes.The expression of small RNAs carried by the endothelial-derived exosomes was detected by high-throughput sequencing and compared with the statically cultured endothelial cellderived exosomes to identify differentially expressed miRNAs.Results In this study,exosomes were successfully isolated from vascular endothelial cells,and disk-shaped vesicles with a diameter of 40~100 nm were observed under transmission electron microscopy.Sequencing of miRNAs carried by exosomes revealed that endothelial cells released a large number of differentially expressed miRNAs via exosomes upon exposure to bacterial lipopolysaccharide.Conclusion The expression profile of miRNAs carried by vascular endothelialderived exosomes changed in the early stage of sepsis,and these differentially expressed miRNAs might play an important role in the pathophysiology of cardiovascular system in the early stage of sepsis.
