基于网络药理学探讨葛根芩连汤抗非酒精性脂肪性肝炎的作用机制

摘要 目的采用网络药理学的方法探究葛根芩连汤(GGQLT)抗非酒精性脂肪性肝炎(NASH)的作用机制。方法采用中药系统药理学数据库与分析平台(TCMSP)、DisGeNET数据库筛选葛根、黄芩、黄连、甘草的活性成分及靶点;利用韦恩图在线软件获得GGQLT抗NASH的靶点;使用Cytoscape3.7.2制作蛋白质-蛋白质相互作用(PPI)网络;利用注释、可视化和集成发现数据库(DAVID)行基因本体(GO)富集及京都基因与基因组百科全书(KEGG)信号通路分析。结果获得GGQLT成分69个,靶点254个,排名前3的成分为槲皮素、谷甾醇、小檗碱。获得NASH靶点434个,GGQLT抗NASH靶点66个。可视化分析显示,GGQLT抗NASH靶点中排名前5的为肿瘤蛋白53(TP53)、V-JUN肉瘤病毒17癌基因同源物(JUN)、原癌基因c-Fos(FOS)、白细胞介素-6(IL-6)、表皮生长因子受体(EGFR)。GO富集分析得到587个条目,KEGG信号通路分析得到116条信号通路。结论本研究通过网络药理学获取GGQLT化学成分及靶点,筛选出GGQLT抗NASH的关键成分及关键靶点,并分析GGQLT抗NASH的作用机制,为GGQLT抗NASH的进一步研究提供一定的理论基础。 Objective To explore the mechanism of Gegen Qinlian Tang(GGQLT)against non-alcoholic steatohepatitis(NASH)by network pharmacology.Methods Traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and DisGeNET database were used to screen the active components and targets of Radix Puerariae,Radix Scutellariae,Rhizoma Coptidis and Radix Glycyrrhizae;the target of GGQLT against NASH was obtained by Wayne diagram online software;the protein-protein interaction(PPI)network was constructed using Cytoscape3.7.2;gene ontology(GO)enrichment and Kyoto encyclopedia of genes and genomes(KEGG)signal pathway analysis were performed using the database for annotation,visualization and integrated discovery(DAVID).Results A total of 69 GGQLT components and 254 targets were obtained,and the top three components were quercetin,sitosterol and berberine.A total of 434 NASH targets and 66 GGQLT anti-NASH targets were obtained.Visual analysis showed that the top 5 GGQLT anti-NASH targets were tumor protein 53(TP53),V-JUN sarcoma virus 17 oncogene homolog(JUN),proto-oncogene c-Fos(FOS),interleukin-6(IL-6)and epidermal growth factor receptor(EGFR).GO enrichment analysis obtained 587 items,and KEGG signaling pathway analysis obtained 116 signaling pathways.Conclusion In this study,the chemical components and targets of GGQLT were obtained through network pharmacology,and the key components and key targets of GGQLT against NASH were screened out,and the mechanism of GGQLT against NASH was analyzed,which provided a theoretical basis for the further study of GGQLT against NASH.
