囊胚玻璃化冷冻对小鼠及其子一代的行为学影响与发育评估 被引量:1

The effect of vitrification of blastula on behaviors and developments in mice and their first filial generations

摘要 目的:观察C57BL/6小鼠囊胚的玻璃化冷冻对复苏移植后连续两代小鼠在生理发育、行为学特征以及生殖力方面的影响,探讨玻璃化冷冻技术的长期安全性。方法:超排卵4周龄C57BL/6雌鼠,与雄鼠交配后收集囊胚进行玻璃化冷冻,观察冷冻囊胚复苏移植出生的小鼠(冻胚组)和子一代小鼠生长发育、行为学特征和生殖力,以新鲜囊胚移植出生的小鼠为对照(鲜胚组)。结果:冻胚组和鲜胚组小鼠在生长发育、学习记忆、运动平衡、社交能力、生殖力及各器官发育上差异均无统计学意义(均P>0.05),两组的子一代各项检测指标差异亦无统计学意义(P>0.05)。结论:小鼠囊胚玻璃化冷冻对子代生长发育和行为没有明显影响,可作为辅助生殖的安全胚胎冷冻方式。 Objective:To investigate the effects of blastocysts vitrification on physiological development,behavioral characteristics and fertility in C57BL/6 mice and their two consecutive generations,and to address the long-term safety of vitrification freezing technology.Methods:4-week-old C57BL/6 female mice with superovulation were mated with male mice and then collected blastocysts for vitrification.The mice that came from the cryopreservation embryos(frozen embryo group)and their offspring were used to evaluate the effect of vitrification on the growth and development,behavioral characteristics and fertility compared with the mice born from fresh blastocyst transplantation(fresh embryo group).Results:There were no significant differences in growth and development,learning and memory,motor balance,social ability,fertility and organ development between frozen embryo group and fresh embryo group(P>0.05).In addition,no statistical distinctions were found in the above indicators in their offspring(P>0.05).Conclusion:Vitrification of blastocysts has no markedly impact on the growth and development as well as behavior of the offspring in mice,suggesting that it could be used as a safe embryo freezing way for assisted reproduction.
