摘要
目的探讨lncRNA MALAT1/miR-125b-5p/程序性死亡因子4(PDCD4)分子轴在心力衰竭患者中的作用机制及与心功能的关系。方法选择2022年7月至2023年4月收治的心力衰竭患者78例为观察组;根据美国纽约心脏病学会(NYHA)分级将患者分为Ⅱ级组、Ⅲ级组、Ⅳ级组;选择同期健康体检者67名为对照组;采用实时荧光PCR法测定两组血液中LncRNA MALAT1、miR-125b-5p和PDCD4表达水平;采用心脏彩色多普勒检测仪(型号APYJG620,购自于美国ATL公司)检测患者左心室舒张末期容积(LVEDV)、室壁运动积分指数(WMSI)、左心室收缩末期容积(LVESV)及左室射血分数(LVEF)水平;采用Pearson相关性分析软件对lncRNA MALAT1/miR-125b-5p/PDCD4分子轴与患者心功能进行相关性分析。结果观察组LncRNA MALAT1、miR-125b-5p mRNA高于对照组(P<0.05);PDCD4 mRNA水平低于对照组(P<0.05);Ⅳ级组LncRNA MALAT1、miR-125b-5p mRNA高于Ⅱ级组和Ⅲ级组(P<0.05);PDCD4 mRNA水平低于Ⅱ级组和Ⅲ级组(P<0.05);观察组心力衰竭患者LVEDV及LVESV水平高于对照组(P<0.05);WMSI及LVEF水平低于对照组(P<0.05);Pearson相关性结果表明,心力衰竭患者LncRNA MALAT1、miR-125b-5p与LVEDV和LVESV呈正相关性(P<0.05);与WMSI和LVEF呈负相关性(P<0.05);而PDCD4与LVEDV和LVESV呈负相关性(P<0.05);与WMSI和LVEF呈正相关性(P<0.05)。结论lncRNA MALAT1/miR-125b-5p/PDCD4分子轴能直接参与心力衰竭的发生及发展,且与心功能存在强相关性。
Objective To explore the mechanism of action of lncRNA MALAT1/miR-125b-5p/Programmed Death Factor 4(PDCD4)molecular axis in patients with heart failure and its relationship with cardiac function.Methods From July 2022 to April 2023,78 patients with heart failure were selected as the observation group;According to the New York Heart Association(NYHA)classification,patients are divided into ClassⅡ,Ⅲ,andⅣgroups;67 healthy individuals who underwent physical examination during the same period were selected as the control group.The expression levels of LncRNA MALAT1,miR-125b-5p,and PDCD4 in the blood of two groups were measured using real-time fluorescence PCR method.The left ventricular end diastolic volume(LVEDV),wall motion integral index(WMSI),left ventricular end systolic volume(LVESV),and left ventricular ejection fraction(LVEF)levels were measured using a cardiac color Doppler detector(model:APYJG620,purchased from ATL Company in the United States).Pearson correlation analysis software was used to analyze the correlation between lncRNA MALAT1/miR-125b-5p/PDCD4 molecular axis and patient cardiac function.Results The LncRNA MALAT1 and miR-125b-5p mRNA in the observation group were higher than those in the control group(P<0.05).The level of PDCD4 mRNA was lower than that of the control group(P<0.05).The levels of LncRNA MALAT1 and miR-125b-5p mRNA in theⅣgroup were higher than those in theⅡandⅢgroups(P<0.05).The level of PDCD4 mRNA was lower than that of the gradeⅡandⅢgroups(P<0.05).The levels of LVEDV and LVESV in patients with heart failure in the observation group were higher than those in the control group(P<0.05).The levels of WMSI and LVEF were lower than those of the control group(P<0.05).Pearson correlation results showed that there was a positive correlation between LncRNA MALAT1,miR-125b-5p,LVEDV,and LVESV in patients with heart failure(P<0.05).There was a negative correlation with WMSI and LVEF(P<0.05),PDCD4 showed a negative correlation with LVEDV and LVESV(P<0.05),there was a positive correlation with WMSI and LVEF(P<0.05).Conclusions The lncRNA MALAT1/miR-125b-5p/PDCD4 molecular axis can directly participate in the occurrence and development of heart failure,and has a strong correlation with cardiac function.
