摘要
目的探讨环状RNA(hsa_circ_0000741)对乳腺癌癌细胞增殖、迁移和侵袭的影响及分子机制。方法通过R语言limma包分析GSE182471和GSE165884数据集;通过常规培养乳腺癌细胞系MCF-7、MDA-MB-231和正常细胞系MCF-10A,提取细胞总RNA,逆转录后进行qRT-PCR测定目的基因表达量;通过lipo2000转染si-hsa_circ_0000741对目的基因进行敲减,将其编为si-hsa_circ_000074组,转染si-nc后将其编为si-nc组,并用qRT-PCR实验检验转染效率,之后进行集落和CCK-8实验对于si-hsa_circ_000074组和si-nc组之间的细胞增殖速率进行检验;开展划痕和侵袭实验对于si-hsa_circ_000074组和si-nc组之间的细胞迁移和侵袭能力进行进行检验。结果GSE182471和GSE165884数据集取交集得出has_circ_0000741在两个数据集中癌组织组均显著上调;qRT-PCR实验发现hsa_circ_0000741在正常细胞系中表达量低于癌细胞系,在三阴性细胞系MDA-MB-231较MCF-7更为显著;集落和CCK-8实验显示si-hsa_circ_000074组的增殖速率明显低于si-nc组;划痕和侵袭实验表明si-hsa_circ_000074组的迁移和侵袭能力明显弱于si-nc组。结论Hsa_circ_0000741在乳腺癌中显著高表达,下调其表达抑制乳腺癌细胞增殖、迁移和侵袭。
Objective To investigate the effect of ring-like RNA(hsa_circ_0000741)on the proliferation,migration and invasion of breast cancer cells and the molecular mechanism.Methods Total cellular RNA was extracted by conventional culture of breast cancer cell lines MCF-7 and MDA-MB-231 and normal cell line MCF-10A,and the expression of target genes was determined by qRT-PCR after reverse transcription,and the target genes were knocked down by transfection with si-hsa_circ_0000741,which was named as si-hsa_circ_0000741 group,and after transfection with si-nc,they were named as si-nc group,and the transfection efficiency was examined by qRT-PCR,followed by colony and CCK-8 assays to examine the cell proliferation rate between si-hsa_circ_0000741 and si-nc groups,and scratch and invasion assays to examine the cell proliferation rate between si-hsa_circ_0000741 and si-nc groups.The cell migration and invasion ability between the si-hsa_circ_0000741 and si-nc groups were examined.Results Analysis of the GSE182471 and GSE165884 datasets taking intersections yielded that hsa_circ_0000741 was significantly upregulated in the cancer tissue group in both datasets.qRT-PCR experiments revealed that hsa_circ_0000741 was less expressed in normal cell lines than in cancer cell lines,and in the triple-negative cell line MDA-MB-231 than MCF-7 was more significant,colony and CCK-8 assays showed that the proliferation rate of si-hsa_circ_0000741 group was significantly lower than that of si-nc group,and scratch and invasion assays showed that the migration and invasion ability of si-hsa_circ_0000741 group was significantly reduced compared with si-nc group.Conclusion Hsa_circ_0000741 was significantly highly expressed in breast cancer,and down-regulation of its expression inhibited proliferation,migration and invasion of nodal breast cancer cells.
