摘要
目的探讨眼睑基底细胞癌组织中SIRT1、JAK2的表达及其临床意义。方法选取经病理检查证实为眼睑基底细胞癌且经手术切除保留的癌组织(70例)作为观察组,另将其同期切除的癌旁组织(70例)作为对照组。采用免疫组化法检测沉默信息调节因子1(SIRT1)、酪氨酸蛋白激酶-2(Janus kinase-2,JAK2)阳性表达率,并分析SIRT1、JAK2表达与眼睑基底细胞癌临床病理特征的相关性。结果眼睑基底细胞癌组织的SIRT1、JAK2表达阳性率明显高于癌旁组织(P<0.05);眼睑基底细胞癌SIRT1高表达者年龄、PCNA增殖指数、肿瘤直径、病理分型与SIRT1低表达者比较存在差异(P<0.05);眼睑基底细胞癌组织JAK2低表达和JAK2高表达者TNM分期比较存在差异(P<0.05);SIRT1表达与眼睑基底细胞癌患者年龄、增殖细胞核抗原(PCNA)增殖指数、肿瘤直径、病理分型呈正相关性;JAK2表达与眼睑基底细胞癌TNM分期呈正相关性(P<0.05)。结论SIRT1与JAK2在眼睑基底细胞癌组织中均呈高表达,与患者年龄、肿瘤直径、TNM分期等临床特征有关,可作为鉴别眼睑基底细胞癌恶性程度的潜在生物标志物。
Objective To investigate the expression and clinical significance of SIRT1 and JAK2 in eyelid basal cell carcinoma tissue.Methods 70 cases of eyelid basal cell carcinoma confirmed by pathological examination and preserved after surgical resection were selected as the observation group,and 70 cases of adjacent cancerous tissue removed at the same time were selected as the control group.Immunohistochemical method was used to detect the positive expression rates of silent information regulatory factor 1(SIRT1)and tyrosine protein kinase 2(JAK2),and the correlation between the expression of SIRT1 and JAK2 and the clinical pathological characteristics of eyelid basal cell carcinoma was analyzed.Results The positive rates of SIRT1 and JAK2 expression in eyelid basal cell carcinoma tissues were significantly higher than those in adjacent tissues(P<0.05);There were differences in age,PCNA proliferation index,tumor diameter,pathological classification between patients with high expression of SIRT1 in eyelid basal cell carcinoma and those with low expression of SIRT1(P<0.05);There was a difference in TNM staging between individuals with low expression of JAK2 and those with high expression of JAK2 in eyelid basal cell carcinoma tissue(P<0.05);The expression of SIRT1 is positively correlated with age,proliferating cell nuclear antigen(PCNA)proliferation index,tumor diameter,and pathological classification in patients with eyelid basal cell carcinoma;The expression of JAK2 is positively correlated with TNM staging of eyelid basal cell carcinoma(P<0.05).Conclusion SIRT1 and JAK2 are both highly expressed in eyelid basal cell carcinoma tissues,which are related to clinical characteristics such as patient age,tumor diameter,TNM staging,and can serve as potential biomarkers for distinguishing the malignancy of eyelid basal cell carcinoma.
