金花茶花的指纹图谱建立、成分鉴定及体外抗氧化活性研究 被引量:2

Establishment of fingerprint,component identification,and in vitro antioxidant activity of Camellia nitidissima

摘要 目的 建立金花茶花的高效液相(HPLC)指纹图谱,对其化学成分进行鉴定,并考察其体外抗氧化能力。方法70%乙醇溶液回流法提取10批次金花茶花,采用Agilent Eclipse Plus C18(250 mm×4.6 mm,5μm)色谱柱,梯度洗脱,建立金花茶花指纹图谱,应用HPLC-MS法分析其化学成分,并用1,1-二苯基-2-苦基肼自由基(DPPH)和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)法评价金花茶花的抗氧化能力。结果 建立金花茶花共有HPLC色谱图,标定共有峰11个,经过中药色谱指纹图谱相似度评价软件(2012版)计算,相似度均在0.98以上。HPLC-MS分析结果显示,金花茶花中成分多为黄酮类化合物。抗氧化能力检测结果显示,金花茶花对DPPH自由基和ABTS自由基均具有较好的清除能力,并呈浓度相关性。结论 建立了金花茶花药材的指纹图谱,其含有丰富的黄酮类化合物,具有良好的体外抗氧化作用。 Objective To establish a HPLC fingerprint of Camellia nitidissima Chi,analyze its chemical components,and study its antioxidant capacity.Methods Extraction of ten batches of Camellia nitidissima Chi were made by refluxing with 70%ethanol solution.Agilent Eclipse Plus C18(250 mm×4.6 mm,5μm)Chromatographic column were used,gradient elution,to establish a fingerprint of Camellia nitidissima Chi,analysis of its chemical components using HPLC-MS,and evaluation of the antioxidant capacity of Camellia nitidissima Chi using DPPH and ABTS methods.Results The HPLC chromatogram of Camellia nitidissima Chi was established,and 11 peaks were calibrated.The similarity was all above 0.98 through calculation of TCM chromatographic fingerprint similarity evaluation software(2012 edition).Meanwhile,HPLC-MS analysis showed that the components of Camellia nitidissima Chi were mostly flavonoids,and the antioxidant capacity of Camellia nitidissima Chi was tested.The results showed that it had good antioxidant capacity to DPPPH free radicals and ABTS free radicals in vitro,and showed a certain concentration dependence.Conclusion We had established a fingerprint of Camea nitidissima Chi,which was rich in flavonoids and had good in vitro antioxidant effects.
