基于网络药理学的康复新液治疗胃溃疡的机理研究 被引量:3

摘要 目的 利用网络药理学研究方法,探索康复新液治疗胃溃疡的机制。方法 通过文献检索收集康复新液的有效化学成分。基于PubChem、Super-PRED、OMIM、GeneCards等数据库,得到有效化学成分作用靶点及胃溃疡相关的疾病靶点,利用韦恩图得到康复新液治疗胃溃疡的相关靶点。利用蛋白-蛋白相互作用网络(PPI)对靶点关系进行预测并通过Cytoscape软件中的“Cytohubba”插件进行关键基因的筛选。最后利用R语言“clusterprofiler”包对靶点进行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析预测康复新液治疗胃溃疡的作用机制。结果 通过文献检索共收集到12种康复新液的有效化学成分,基于各个数据库获得了134个康复新液治疗胃溃疡相关靶点,根据PPI网络分析及“Cytohubba”插件筛选出13个靶点,分别为:STAT3、HSP90AA1、MTOR、MMP9、EZH2、CXCR4、MMP2、PTGS2、TLR4、CDK2、PTPN11、GSK3B、PDGFRB。GO分析表明:靶基因在生物过程中主要集中在MAPK级联正调控、对氧气水平的反应、对氧化应激的反应、细胞对氧化应激的反应、细胞因子产生的正向调节等;KEGG分析表明:靶基因主要集中在JAK-STAT、PI3K-Akt、HIF-1、MAPK信号通路、EGFR酪氨酸激酶抑制剂耐药性、T辅助细胞17细胞分化等。结论 康复新液对胃溃疡的治疗涉及多个靶点以及多种途径。主要以抗氧化应激及抗炎症反应为主。JAK-STAT、PI3K-Akt、MAPK信号通路可能是康复新液治疗胃溃疡的关键通路。 【Objective】To explore the mechanism of KANGFUXIN liquid in the treatment of gastric ulcer by network pharmacology.【Methods】The effective chemical constituents of KANGFUXIN liquid were collected by literature retrieval.Based on PubChem,Super-Pred,OMIM,GeneCards and other databases,the targets of effective chemical components and disease targets related to gastric ulcer were obtained,and the targets related to the treatment of gastric ulcer were obtained by using Venn diagram.Protein-protein interaction(PPI)network was used to predict target relationships and key genes were screened by"Cytohubba"in Cytoscape.Finally,GO and KEGG enrichment analysis were performed on the targets using R language"clusterprofiler"package to predict the mechanism of KANGFUXIN liquid in treating gastric ulcer.【Results】A total of 12 effective chemical components of KANGFUXIN liquid were collected through literature search,134 targets related to the treatment of gastric ulcer were obtained based on various databases,and 13 targets were screened out according to PPI network analysis and"Cytohubba",which were as follows:STAT3,HSP90AA1,MTOR,MMP9,EZH2,CXCR4,MMP2,PTGS2,TLR4,CDK2,PTPN11,GSK3B,PDGFRB.GO analysis showed that target genes mainly focused on positive regulation of MAPK cascade,response to oxygen levels,response to oxidative stress,cellular response to oxidative stress,positive regulation of cytokine production,etc.KEGG analysis showed that target genes mainly concentrated in JAK-STAT,PI3K-Akt,HIF-1,MAPK signaling pathway,EGFR tyrosine kinase inhibitor resistance,Th17 cell differentiation,etc.【Conclusion】The treatment of gastric ulcer with KANGFUXIN liquid involves multiple targets and pathways mainly to anti-oxidative stress and anti-inflammatory response.JAK-STAT,PI3K-Akt and MAPK signaling pathways may be the key pathways of KANGFUXIN liquid in the treatment of gastric ulcer.
