摘要
目的观察针刺对慢性阻塞性肺疾病(COPD)大鼠肺组织中硫化氢(H_(2)S)含量及其合成酶胱硫醚β-合酶(CBS)、胱硫醚γ裂解酶(CSE)表达的影响。方法选取24只6周龄清洁级雄性SD大鼠,体重(200±10)g,按随机数字表法将其分为对照组、模型组、电针组,每组8只。对照组常规饲养,模型组和电针组采用香烟烟雾联合脂多糖法构建COPD模型。造模成功后,电针组采用电针针刺双侧肺俞和足三里,连续2周,模型组和对照组不做处理。干预结束后,检测三组用力肺活量(FVC)、第0.1秒用力呼气量(FEV_(0.1))、FEV_(0.3)、呼气流量峰值(PEF)。观察三组肺部病理变化,采用酶联免疫吸附试验检测三组肺组织和血浆白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平,检测肺组织H_(2)S、CBS、CSE含量,采用蛋白质印迹法观察三组肺组织CBS、CSE蛋白表达,进一步分析肺组织IL-1β、TNF-α与H_(2)S的相关性。结果模型组FEV_(0.1)、FEV_(0.3)、FEV_(0.1)/FVC、FEV_(0.3)/FVC、PEF均低于对照组,差异有统计学意义(P<0.05),其肺部病理结果显示炎症细胞浸润,肺间质增厚。模型组肺组织及血浆IL-1β、TNF-α均高于对照组,肺组织H_(2)S、CBS、CSE含量及CBS、CSE蛋白表达均高于对照组,差异有高度统计学意义(P<0.01)。电针组FEV_(0.1)、FEV_(0.3)、FEV_(0.1)/FVC、FEV_(0.3)/FVC、PEF均高于模型组,差异有统计学意义(P<0.05),其肺部病理表现减轻。电针组肺组织及血浆IL-1β、TNF-α均低于模型组,肺组织H_(2)S、CBS、CSE含量及CBS、CSE蛋白表达低于模型组,差异有统计学意义(P<0.05)。相关性分析结果显示,大鼠肺组织IL-1β、TNF-α与H_(2)S呈正相关(r>0,P<0.01)。结论电针针刺肺俞和足三里可下调COPD大鼠肺组织H_(2)S及CSE、CBS含量及表达,H_(2)S可能参与针刺的抗感染作用。
Objective To observe the effects of acupuncture on the content of hydrogen sulfide(H_(2)S)and the expression of cystathionine-β-synthase(CBS)and cystathionine-γlyase(CSE)in lung tissue of chronic obstructive pulmonary disease(COPD)rats.Methods A total of 246-week-old clean grade male SD rats with body weight of(200±10)g were selected,and divided into control group,model group,and electroacupuncture group by random number table method,with eight rats in each group.The control group was fed conventionally.The COPD model was established by cigarette smoke combined with lipopolysaccharide in model group and electroacupuncture group.After successful modeling,the electroacupuncture group was treated with electricacupuncture on both fèishùand zúsānlǐfor two consecutive weeks,while the model group and control group were not treated.After the intervention,forced vital capacity(FVC),forced expiratory volume in 0.1 second(FEV_(0.1)),FEV_(0.3),and peak expiratory flow(PEF)were measured.The pathological changes in the lungs of the three groups were observed.The levels of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in lung tissue and plasma of the three groups were detected by enzyme-linked immunosorbent assay.The contents of H_(2)S,CBS,and CSE in lung tissue were detected.The correlation between IL-1βand TNF-αin lung tissue and H_(2)S was further analyzed.Results FEV_(0.1),FEV_(0.3),FEV_(0.1)/FVC,FEV_(0.3)/FVC,and PEF in the model group were lower than those in the control group,and the differences were statistically significant(P<0.05).Its pulmonary pathological results showed inflammatory cell infiltration and pulmonary interstitial thickening.IL-1βand TNF-αin lung tissue and plasma in model group were higher than those in control group,while H_(2)S,CBS,CSE contents,and CBS,CSE protein expressions in lung tissue were higher than those in control group,and the differences were highly statistically significant(P<0.01).FEV_(0.1),FEV_(0.3),FEV_(0.1)/FVC,FEV_(0.3)/FVC,and PEF in electroacupuncture group were higher than those in model group,and the differences were statistically significant(P<0.05).IL-1βand TNF-αin lung tissue and plasma in the electroacupuncture group were lower than those in the model group,while the H_(2)S,CBS,CSE contents and the CBS and CSE protein expression in lung tissue were lower than those in the model group,the differences were statistically significant(P<0.05).Correlation analysis showed that IL-1β,TNF-α,and H_(2)S in lung tissue were positively correlated(r>0,P<0.01).Conclusion Electroacupuncture in fèishùand zúsānlǐcan down-regulate the content and expression of H_(2)S,CSE,and CBS in lung tissue of COPD rats,and H_(2)S may be involved in the anti-infection effect of acupuncture.
