基于罗丹明B酰肼的化合物铜离子络合能力测定的新方法 被引量:1

摘要 目的:开发一种以罗丹明B酰肼为探针的铜离子络合剂分析方法。方法:通过合成罗丹明B酰肼来监测游离铜离子,罗丹明B在铜离子的作用下可发生开环反应,其荧光强度可用于检测铜离子,以此来评价化合物与铜离子的络合能力从而筛选铜络合剂,并对影响该测定方法的各个反应条件进行优化。结果:以pH为7的乙腈-水溶液为溶剂,铜离子与罗丹明B酰肼反应时间大于8 min时,可观察到稳定的荧光信号,从而比较了化合物的络合能力。以典型的铜离子络合剂曲酸和EDTA为模型化合物,验证了该分析方法的可行性。结论:本研究开发的方法能够简单快速地评价化合物的铜离子络合能力,为神经退行性疾病潜在药物的筛选提供新的方法。 Objective:Develop a method for analysis of copper ion chelators using rhodamine B hydrazide as a probe.Methods:Rhodamine B hydrazide was synthesized to monitor free copper ions in this study.Rhodamine B can undergo ring-opening reaction in the presence of copper ions,and the resulting fluorescence intensity can be used to detect copper ions for assessing the copper chelating ability of compounds to screen copper chelators.The reaction conditions affecting the method were optimized.Results:In acetonitrile-water solution at pH=7,a stable fluorescence signal was observed when the reaction time between copper ions and rhodamine B hydrazide exceeded 8 min,allowing for comparison of the chelating abilities of compounds.The feasibility of this analytical method was validated using the typical copper chelators kojic acid and EDTA as model compounds.Conclusion:The developed method in this study can rapidly and simply assess the copper ion chelating abilities of compounds,thus providing a new approach for screening potential drugs for neurodegenerative diseases.
